SAM-P8小鼠痕迹性眨眼条件反射的获得及其PFC-ERP变化

摘  要:目的:研究快速老化P8小鼠(senescence accelerated mouse-prone/8,SAM-P8)痕迹性眨眼条件反射(trace eyeblink conditioning,TEBC)的获得及其前额叶(prefrontal cortex,PFC)事件相关电位(event related potential,ERP)的变化。方法:实验动物为SAM-P8组(n=11)与对照小鼠SAM-R1(senescence accelerated mouse-resistant/1,SAM-R1)组(n=11);对2组小鼠进行条件刺激(conditioned stimulus,CS)与非条件刺激(unconditioned stimulus,US)配对的经典TEBC训练,其中CS为250 ms、3 k Hz、85 d B的纯音,US为强度为3 Psi、持续时间100 ms的束状氧气对动物角膜进行给气。观察2组动物的经典TEBC的条件反应(conditioned response,CR)的习得及其PFC-ERP的变化。结果:行为学结果方面,SAM-P8可以获得有限的经典TEBC(CS:250ms 85 d B的纯音),SAM-R1的条件反应发生率(incidence rate of conditioned response,CR%)明显高于SAM-P8(P<0.05)。PFCERP结果方面:在CR期,无论是峰峰幅度和峰潜伏期未见SAM-P8与SAM-R1 2组间有明显差别,且2组组内训练各期之间没有统计学差异(P>0.05);在CR前期,SAM-P8的PFC-ERP的峰峰幅度显著低于SAM-R1(P<0.01),SAM-R1行为习得中期PFC-ERP的峰峰幅度明显高于其行为习得早期(P<0.05),但SAM-P8未见此显著变化。结论:SAM-P8小鼠对经典TEBC的学习能力有损伤但并不完全;在TEBC建立过程中,SAM-P8小鼠其前额叶的参与程度低于SAM-R1小鼠,PFC可能更多参与TEBC的中期获得过程,但这一时间特征对于SAM-P8似乎并不明显。Objective : To investigate the changes of the learning of trace eyeblink conditioning (TEBC) and prefrontal cortex (PFC)' s event-related potential in senescence accelerated mouse-prone/8(SAM-PS) mice. Methods:Experimental animals were divided into SAM-P8 group(n=11 ) and SAM-R1 control group(n=1 1);two groups received conditioned stinmlus(CS) and unconditioned stimulus (US) paired training,in which,CS is a 250 ms,3 kHz,85 dB tone and US is a 3.0 Psi, 100 ms corneal air puff. Then the CR acquire- ment of TEBC in the two groups were observed,and changes of PFC's event-related potential during this process were compared be- tween the two groups. Results:Results behavioral results :SAM-P8 mice acquired limited classical TEBC with optimal CS(250 ms, 3 kHz, 85 dB tone) ;the CR% of SAM-R1 was higher than that of SAM-P8 (P〈0.05). In PFC-ERP:there are no significant difference in peak-peak amplitude or latency of peak between SAM-P8 group and SAM-R1 group,and there was also no significant difference in training periods between two groups(P〉0.05) in CR period. In pre-CR period:peak-peak amplitude of PFC-ERP in SAM-P8 mice reduced observably compared with that of SAM-R1 (P〈0.05) ;peak-peak amplitude of PFC-ERP in medium-term of behavior training was much higher than that in early period of training in SAM-R1 mice (P〈0.05) ,but no related changes in SAM-P8 were observed (P〉0.05). Conclusion:SAM-PS's learning capability of TEBC has been impaired but the impairment is incomplete. During the es-tablishment of TEBC,PFC involvement of SAM-P8 mice is less than that of SAM-R1 mice;PFC may be more involved during the medium-tema of behavior training,but this time characteris- tic is not obvious in SAM-P8.
