摘  要:为探究固定化细胞对三氯乙烯(TCE)的生物降解特性及机理,以优选的活性炭纤维为固定化材料,构建双塞头固定化细胞反应器;利用填埋场覆盖层富集筛选的高效混合菌SWA1为生物介质开展细胞固定化及TCE生物降解研究;运用扫描电镜,荧光定量PCR和高通量测序技术分别考察TCE降解前后固定化微生物微观结构、关键酶表达丰度和混合菌群落结构变化.结果显示:(1)该固定化细胞反应器对微生物具有良好的固定化效果,且微生物可维持较高活性(甲烷氧化速率为0.6μmol g^(-1) d^(-1));(2)在TCE初始浓度为18 mg/L时,反应1 d降解率为94.1%,降解过程中有8.7μmol有机氯转化为氯离子;(3)反应器中TCE降解关键酶为甲烷单加氧酶和苯酚羟化酶,TCE降解后优势菌属为Anaerolineaceae_unclassified(0.6%-19.9%)和甲基孢囊菌属Methylocystis(0-15.8%).该双塞头固定化细胞反应器能高效去除氯代烃类污染物,可为含氯有机废水高效生物治理的工程应用提供参考.An experiment on trichloroethene (TCE) biodegradation in a reactor with immobilized cells was performed to explore the characteristics and mechanism of TCE biodegradation by an immobilized mixed culture of SWA1, which was isolated from landfill-cover soil. Activated carbon fiber was selected for immobilization, and a reactor with two openings was developed and set up. Then, SWA1 was continuously enriched in the reactor, which was filled with activated carbon fiber. TCE was added to the reactor, and then, the rates of TCE degradation and Cl- production were monitored. The microstructure of the immobilized microorganisms was analyzed by scanning electron microscopy, the relative abundance of key enzyme genes was analyzed by real-time quantitative PCR, and the microbial community structure within the reactor was analyzed by MiSeq sequencing before and after biodegradation. The results showed that: 1) Immobilized microorganisms at high density existed in the reactor, and the rate of CH4 oxidation was 0.6 μmol g-1 d-1 in the reactor. 2) The efficiency of TCE degradation was 94.1% in one day at initial TCE concentration of 18 mg/L; meanwhile, organochlorine at 8.7 μmol was converted to chloride. 3) The key enzymes were methane monooxygenase and phenol hydroxylase. The dominant bacteria in the genus were Anaerolineaceae_unclassified (0.6% to 19.9%) and Methylocystis (0 to 15.8%) after TCE biodegradation. The reactor with two openings with immobilized cells can effectively remove chlorinated hydrocarbon pollutants, and can provide information for engineering applications in the high-efficiency biological treatment of chlorinated organic wastewater.
