母鼠注射海洛因对仔鼠大脑额叶结构及相关蛋白表达的影响

摘　　要：研究母鼠注射海洛因对仔鼠大脑额叶结构与相关蛋白表达的影响,选20只母鼠随机分为空白对照组和海洛因1,2,3组,从受孕第10d开始,各孕鼠分别腹腔连续注射生理盐水和2.0,4.0,6.0g·L-1的海洛因溶液,直至孕鼠分娩后15d,于仔鼠出生5d,10d和15d分别称量检测仔鼠大脑的重量,用光学显微镜观察其大脑额叶的显微结构变化,用免疫组化法检测额叶皮层NGF和Caspase-3蛋白的表达.结果表明,各海洛因组仔鼠大脑重量均显著或极显著低于空白对照组(P<0.05或P<0.01);海洛因组仔鼠额叶皮层组织结构有不同程度的发育缺陷,外颗粒层、内颗粒层、锥体细胞层和多形细胞层的细胞均出现空泡化和细胞凋亡现象,额叶皮层NGF表达增强,Caspase-3蛋白的表达极显著增强(P<0.01).表明母鼠注射海洛因影响仔鼠大脑额叶组织的正常发育.The effects of heroin on the frontal lobe structure and related protein expression in the brain ofyoung mice are studied when pregnant mice injecting heroin. Twenty pregnant mice are randomly dividedinto control group and heroin group 1,2,3. The 10th day of conception, pregnant mice are injected intraperitoneally with normal saline and 2. 0,4. 0, 6. 0 g·L-1 heroin solution until 15 days 5 d,10 d and 15d the changes of brain weight are detected after birth, the changes of frontal lobe structureare observed by biological microscope, and the expression of NGF and Caspase-3 protein in frontal cortex is detected by immunohistochemistry. The results show that the brain weight of the young mice in the heroin groups are significantly or extremely significant lower than that in the control group（P〈0. 05 or P 〈0.01）. The structure of the frontal cortex in the heroin groups has different degrees of damage. External granular layer, internal granular layer, pyramidal layer and polymorphic layer of brain frontalcortex in young mice occur cell vacuolization and cell apoptosis. The expression of NGF in the frontal cortex is increased. The expression of Caspase-3 protein in the frontal cortex is extremely significantly increased（P〈0. 01）. The results show that heroin affects the development and structtissue in young mice when pregnant mice injecting heroin.
