摘  要:目的探讨循环型micro RNA-26a(mi R-26a)及靶基因PTEN在系统性红斑狼疮(SLE)的表达,以及在SLE发生、发展中可能发挥的作用。方法选取36例SLE和10例健康体检者血液标本,实时荧光定量聚合酶链反应检测血清mi R-26a、外周血单个核细胞(PBMC)中PTEN的表达,统计分析mi R-26a、PTEN与SLE临床资料的相关性,利用双荧光素酶报告系统验证PTEN是否为mi R-26a的靶基因。结果 SLE患者血清mi R-26a m RNA的表达高于健康人群(P<0.05),PBMC中PTEN m RNA的表达低于健康人群(P<0.05)。SLE患者mi R-26a、PTEN的表达水平与C3、抗ds-DNA、红细胞沉降率、狼疮活动指数相关。mi R-26a转染Ba F3细胞后,PTEN表达下调,PI3K表达上调,转染组荧光素酶活性下降(P<0.05)。结论 SLE患者血清中过表达的mi R-26a可能通过下调靶基因PTEN,上调PI3K/AKT信号通路,参与SLE的发生、发展。Objective To explore the expressions of circulating mi R-26 a and its target gene PTEN in systemic lupus erythematosus(SLE) and their possible roles in the occurrence and development of SLE.Methods The expressions of circulating mi R-26 a and PTEN in peripheral blood mononuclear cells(PBMCs)were detected by q RT-PCR after blood samples were collected from 36 cases of SLE and 10 healthy people.The correlations between mi R-26 a, PTEN and clinical data of SLE were statistically analyzed. Dual luciferase reporter system was used to test whether PTEN was the target gene of mi R-26 a. Results The expression of mi R-26 a m RNA in the serum of the SLE patients was higher than that in the healthy people(P < 0.05), and the expression of PTEN m RNA in the PBMCs of the SLE patients was lower than that in the PBMCs of the healthy people(P < 0.05). The expression levels of mi R-26 a and PTEN in the patients with SLE were correlated with C3, anti-ds DNA, ESR and SLEDAI. After mi R-26 a was transfected to Ba F3 cells, the expression of PTEN was down-regulated, and the expression of PI3 K was up-regulated, and the luciferase activity of the transfected group was decreased(P < 0.05). Conclusions The over-expression of circulating mi R-26 a may participate in the occurrence and development of SLE through down-regulation of its target gene PTEN and up-regulation of PI3 K/AKT signaling pathway.
