摘  要:甜高粱(Sorghum bicolor L.Beauv)作为重要的能源作物已经为大家所认可。本研究利用集群分离分析(BSA)法结合SRAP分子标记技术相结合,以141个甜高粱"W455"和粒用高粱"忻粱52"杂交形成的重组自交系F2:3群体为材料,寻找与甜高粱含糖量性状相关的SRAP分子标记。结果表明:含糖量遗传属于单基因加显或单基因加性效应模式,出汁率遗传是由多基因共同控制的数量性状;通过SRAP分子标记发现,在7号染色体上找到1个与含糖量基因连锁的SRAP标记M3E7-S248,通过测序比对发现其与高粱腺苷二磷酸葡萄糖焦磷酸化酶亚基SH2有79%的相似度;在6号染色体上找到了3个与出汁率基因连锁的SRAP标记M8E2-J727(M8E2-J712)、M8R12-J241和F13E9-J150,其中M8E2-J712与M8E2-J727为共显性标记;通过测序比对发现标记M8E2-J712与多种植物叶绿体上的氧捕获增强蛋白高度一致;标记M8E2-J727与双色高粱未知功能蛋白m RNA中的一段序列具有99%的一致性;标记M8R12-J241和F13E9-J150均与6号染色体上的未知基因序列具有高度一致性。As an important energy crop, sweet sorghum(Sorghum bicolor L. Beauv) has been recognized by us. In this research we used the recombinant inbred F2:3population derived from the hybridization of 141 sweet sorghum"W455"and grain sorghum "Xinliang 52"as material, located the molecular markers associated with sugar content of sweet sorghum by using BSA method combined with SRAP molecular markers. The results shown that the sugar content belonged to a single gene with additive-dominant effects or single gene with additive effect and the juice yield was controlled by poly-genes. One sugar content of the SRAP marker M3E7-S248 on chromosome7 was found by SRAP molecular markers. From the comparison of sequencing, we found that it had 79% similarity with sorghum adenosine two phosphate glucose pyrophosphorylase subunit SH2, Three of the juice yield SRAP markers linked to juice rate gene were found on chromosome 6 that M8E2-J727(M8E2-J712), M8R12-J241 and F13E9-J150. M8E2-J712 and M8E2-J727 were co-dominant markers. The sequencing showed that M8E2-J712 labeled with a variety of plant chloroplast capture oxygen enhanced protein was highly consistent, consistent sequence of marker M8E2-J727 and double color sorghum protein of unknown function in m RNA was 99%,M8R12-J241 and F13E9-J150 markers were detected on chromosome 6.
