摘  要:目的观察石菖蒲挥发油微乳对低氧诱导SH-SY5Y细胞线粒体损伤的影响,并探讨其作用机制。方法低氧处理SH-SY5Y细胞24 h后分别加入不同浓度的石菖蒲挥发油微乳含药血清,采用MTT法检测细胞存活率,JC-1荧光探针测定法和共聚焦显微镜观察线粒体膜电位,通过实时逆转录聚合酶链反应(real-time RT-PCR)测定细胞中Bcl-2和Bax mRNA的表达。结果石菖蒲挥发油微乳可以显著提高低氧环境下SH-SY5Y细胞的存活率(P<0.01),逆转线粒体跨膜电位的下降,并显著降低细胞中Bax mRNA表达(P<0.01)。结论石菖蒲神经保护的机制可能与线粒体保护、减少细胞凋亡相关。Objective To investigate the effect of serum containing Acorus tatarinowii Schott ( ATS ) on hypoxia-induced mitochondrial injure in SH-SY5Y cells. Methods The SH-SY5Y cells were exposed to hypoxia culture, and the cell viability was measured by the MTT method. The mitochondrial membrane potential was detected by the JC-1 method and the scanning confocal microscopy. The mRNA levels of Bcl-2 and Bax were detected. Results Cell viability experiments revealed that ATS effectively attenuate hypoxia-induced cell death ( P〈0.01) . ATS also reversed decline in the mitochondrial membrane potential induced by hypoxia. The mRNA expression of Bax was significantly decreased by ATS ( P〈0.01) . Conclusion ATS may protect against hypoxia-induced mitochondrial injure in SH-SY5Y cells by recovering the mitochondrial function.
