摘 要:目的建立三叶青藤茎叶药材饮片质量控制方法。方法采用薄层色谱法鉴别药材饮片;建立其砷盐、重金属杂质限量检查法以及水分、总灰分、酸不溶性灰分检测法;测定其浸出物含量;以异荭草苷为指标性成分,采用NaNO_2-Al(NO_3)_3-NaOH比色法,在500nm检测波长、优化显色反应条件,控制三叶青藤茎叶药材饮片中总黄酮含量。结果 3批三叶青藤茎叶药材饮片薄层色谱在对照物出现斑点的相应位置显相同颜色荧光斑点,且各成分分离良好;4批三叶青藤茎叶药材饮片水分测定结果为4.05%~6.77%,总灰分6.06%~7.25%,酸不溶性灰分0.64%~1.65%,水溶性浸出物16.00%~27.09%,醇溶性浸出物15.42%~27.07%,砷盐杂质限量<1ppm,重金属杂质限量<10ppm;建立的比色法在7.73~61.82μg/mL范围内线性关系良好,回归方程A=1.50×10^(-2)C-1.78×10^(-2)(r=0.999 6),4批三叶青藤茎叶药材饮片总黄酮含量20.83~21.50 mg/g。结论所建立的薄层色谱鉴别、水分、总灰分、酸不溶性灰分、浸出物及总黄酮含量测定法可作为三叶青藤茎叶药材饮片内控质量标准的参考依据。Objective To establish the quality control methods of the aerial parts of Tetrastigma Hemsleyanum herbal pieces. Methods Identify herbal pieces by TLC method;The moisture,total ash,acid-insoluble ash and extract were analyzed according to the methods in Chinese Pharmacopoeia(2015).With isoorientin used as reference,the detection wave length set at 500 nm and the color reaction conditions optimized,the content of total flavonoids was determined by NaNO2-Al(NO3)3-NaOH colorimetric method. Results In the TLC conditions,three batches of the aerial parts of Tetrastigma Hemsleyanum herbal pieces showed the same color spots as the isoorientin reference,and the components were well separated. The moisture,total ash,acid-insoluble ash,water-soluble and alcohol-soluble extract of the samples varied in the ranges of 4.05%-6.77%,6.06%-7.25%,0.64%-1.65%,16.00%-27.09% and 15.42%-27.07% respectively. The contents of heavy metals and arsenic salts were less than ten over one million and one over one million respectively. There was a good linear relationship in the range of 7.73-61.82μg/mL,with the equation A=1.50×10^-2C-1.78×10^-2(r=0.999 6). The flavonoids content in four batches of the aerial parts of Tetrastigma Hemsleyanum herbal pieces varied in the range of 20.83-21.50 mg/g. Conclusion The results can be used as a reference to the quality control of the aerial parts of Tetrastigma Hemsleyanum herbal Pieces.
