摘  要:以金纳米溶胶、银纳米溶胶和金-银合金纳米溶胶为基底测定了小鼠血清表面增强拉曼光谱(SERS).观察了血清蛋白质结构的主链、侧链和二级结构的振动峰(958、1 003、1 015、1 146、1 352 cm^(-1)),以及血清中少量碳水化合物D-甘露醇和脂类的特征峰(527、1 469 cm^(-1)),并比较了3种金属溶胶基底的SERS增强效应.结果表明,以金-银合金纳米溶胶为基底的SERS谱峰都得到增强,特别是527、958、1 003、1 146、1 352、1 469 cm^(-1)处信号加强明显.以银纳米溶胶为基底的SERS中蛋白质结构的振动峰也有一定的增强,但是在527 cm^(-1)处只有一小峰,未见1 469 cm^(-1)峰.以金纳米溶胶为基底的SERS中蛋白质结构的振动峰也有所加强,但527、1 469、1 547 cm^(-1)峰消失.实验结果表明,金-银合金纳米溶胶的SERS活性最强,银纳米溶胶其次,金纳米溶胶最差.The surface-enhanced Raman spectroscopy ( SERS) in mice serum was determined with the gold, silver and Au-Ag alloy nano sol as substrates. The characteristic peaks at 958,1 003,1 015,1 146 , 1 352 cm 1 which assigned to the main chain, side chain and secondary structure of serum protein were observed. And the vibrating peaks observed at 527 , 1 469 cm 1 were the characteristic peaks of small a-mount of carbohydrate D-mannitol and lipid in serum. Moreover, the SERS enhancement effects of three kinds of metal sol substrate were compared. The results indicated that SERS peaks were enhanced by u-sing Au-Ag alloy nano sol as substrate. Especially, the characteristic peaks were enhanced significantly at 527 ,958,1 003,1 146,1 352 , 1 469 cm 1. When using silver nano sol as substrate, the SERS character-istic peaks of protein structure had certain enhancement. But merely there was a small peak at 527 cm 1 , the peak of 1 469 cm 1 was invisible. When using gold nano sol as substrate, the characteristic peaks of protein structure were also strengthened, but the peaks at 527,1 469 , 1 547 cm 1 were disappeared. The results showed that the SERS activity of Au-Ag alloy nano sol was the strongest, silver nano sol was mod-erate ,and gold nano sol was the weakest.
