摘 要:目的了解湖南省2015-2016年引起手足口病的CV-A6型肠道病毒病原学特征,为手足口病科学防控提供依据。方法对2015-2016年湖南省手足口病哨点医院和地市级疾控中心上送的其他肠道病毒阳性标本进行CV-A6核酸检测,选择有代表性的标本进行CV-A6 VP1区全长序列扩增和测序。运用Seqman和MEGA 5.2软件对获取序列与从Gen Bank下载的CV-A6 VP1区全长序列进行遗传进化分析。结果共对281份非EV-A71和非CV-A16肠道病毒阳性标本进行了CV-A6核酸检测,142份标本检测结果为CV-A6阳性,阳性率为50.53%,对不同地区和不同年份的有代表性的69份标本进行VP1区全长序列测定,60份测序成功,病毒型别均为CV-A6的D1亚型。结论湖南省存在引起手足口病的非EV-A71和非CV-A16的CV-A6肠道病毒感染,其基因型别为D1亚型,与我国其他地区流行的病毒亚型基本一致。Objective To investigate the pathogenic features of coxsackievirus A6 [ CV-A6) associated with hand, foot and mouth disease (HFMD) in Hunan Province during 2015-2016 so as to provide a basis for scientific prevention and control of HFMD. Methods The positive specimens of other enteroviruses sent by HFMD sentinel hospitals and prefecture-level CDCs in Hunan Province during 2015-2016 were collected to perform CV-A6 nucleic acid detection. CV-A6 real-time PCR positive speci- mens were then subjected to RT-PCR for full length sequence amplification and sequencing of VP1 region. The phylogenetic analy- sis was performed to analyze the full-length gene sequences encoding VP1 of CVA6 isolates and sequences downloaded from Gen- Bank by using Seqman and MEGA 5.2 software packages. Results A total of 281 non-EV-A71 and non-CV-A16 enterovirus positive specimens were detected by CV-A6 real-time PCR assay. 142 specimens were positive for CV-A6, with the positive rate being 50.53%. 69 specimens from different regions and different years were selected for determining the full length sequence of VP1 region and 60 were successfully determined. It was found that all sequences detected belonged to CV-A6 subtype D cluster 1. Conclusions There are non-EV-A71 and non-CV-A16 enterovirus infections caused by CV-A6 in hand, foot and mouth dis- ease in Hunan Province, and the isolates belong to the D1 subgenotype, which is consistent with the prevalent subtypes in other re- gions in China.
