摘  要:目的:建立测定异烟肼HPLC方法,以钴60γ射线(^(60)Co-γ)为辐射源,考察辐射对异烟肼稳定性的影响。方法:采用Diamosil C_(18)(2)色谱柱(250 mm×4.6 mm,5μm);甲醇-0.2%磷酸溶液(2∶98)为流动相,流速:1.0 ml·min^(-1),检测波长:254 nm,进样量:10μl。分别配制不同准确初始浓度的异烟肼溶液(10,50,100,100μg·ml^(-1)+N_2饱和、100μg·ml^(-1)+O_2饱和、100μg·ml^(-1)+10%叔丁醇及异烟肼粉末)等,经不同辐照剂量(0.3,0.6,1,2.5,5,10 k Gy)的^(60)Co-γ辐照后,HPLC法测定溶液中剩余异烟肼的含量。结果:异烟肼的线性范围为1~200μg·ml^(-1)(r=0.999 0)。平均回收率为98.13%,RSD为0.65%。异烟肼的降解率随吸收剂量的增大而增大,随浓度的增大而减小,粉末状态几乎不降解。O_2的存在可明显促进异烟肼的降解,N_2和叔丁醇的存在可以在一定程度上抑制异烟肼的降解。结论:异烟肼水溶液对^(60)Co-γ辐照敏感,其浓度越低,辐照剂量越大,降解越彻底。Objective: To establish an HPLC method for the determination of isoniazid and explore the impact of 60 Co-~, on the stability of isoniazid. Methods: A Diamosil C18 (2) column (250 mm ×4.6 mm,5μm) was used. The mobile phase consisted of methanol and 0.2% phosphoric acid(2:98). The flow rate was 1.0 ml·min-1 , and the detection wavelength was at 254 nm. The isoniazid solutions at different accurate initial concentration (10, 50, 100μg·ml-1, 100μg·ml-1 + saturated N2, 100μg·ml-1 + saturated 02, 100μg·ml-1 + 10% tert-bntanol and isoniazid powder) were irradiated by 60Co-γ at different doses (0.3, 0.6, 1, 2.5, 5, 10 kGy). And then, the concentrations of remained isoniazid were determined by HPLC. Results: Isoniazid was in a good linear relationship within the range of 1-200μg·ml-1 ( r = 0. 999 0) with the recoveriy of 98.13 % (RSD = 0.65% ). The degradation rate of isoniazid was affected by the initial concentration and the radiation dose, which increased with the increase of absorption amount and increased with the concentration increase, and the powder almost had no degradation. The degradation of isoniazid + O2 significantly decreased, and that of isoniazid + N2 and isoniazid + tert-bntanol was inhibied to a certain extent. Conclusion: Isoniazid is sensitive to 60Co-γ irradiation , and its degradation is higher with lower concentration and higher irradiation dose.
