摘　　要：目的了解RhD抗原免疫刺激途径,建立D抗原免疫刺激活化的模型。方法选取RhD血型不同的供者,采用红细胞与单个核细胞直接混合培养,以及DC细胞负载RhD抗原后与单个核细胞混合培养2种实验方案,流式细胞仪检测CD4^+和CD8^+T细胞的活化量。结果 RhD(-)个体的DC细胞负载D抗原后,可以有效活化RhD-个体的CD4^+和CD8^+T细胞,而直接混合培养方式则不能。结论本研究采用DC细胞负载RhD抗原的方法,成功诱导了RhD(-)个体的T细胞活化和增殖,验证了RhD抗原的胸腺依赖性抗原特性,建立了D抗原免疫刺激T细胞活化的模型,可以用于输血免疫的研究。Objective To understand the pathway of RhD antigen immunoreactive,and establish D antigen immunostimulatory activation model.Methods Different donors of RhD blood group were selected,and direct mixed culture with mononuclear cells.Another protocol was mixed with mononuclear cells cultured after using dendritic cells loaded D antigen.Flow cytometry was used to detect the activation of CD4~＋and CD8~＋T cells.Results Using DC cells from RhD-individuals to load D antigen could effectively activate the RhD-individual CD4~＋and CD8~＋T cells,and direct mixing culture was invalid.Conclusion In this study,DC cells loaded with RhD antigen were successfully to induce the activation and proliferation of T cells from RhD-individuals.RhD antigens were confirmed to be thymus-dependent antigens,and the model of D antigen activation T cells was established which could be used in the study on transfusion immunity.
