摘  要:目的评估血液制品中甘油、上清蛋白、血浆蛋白、游离Hb以及亚甲蓝检测试剂盒的可行性并建立室内质量控制。方法选择市售生化试剂盒,自制室内质控品并制定质量控制范围,分别用分光光度计和生化仪器同时对血液制品中的甘油、上清蛋白、血浆蛋白、游离Hb以及亚甲蓝检测进行检测,甘油检测采用GPO-PAP酶比色法,上清蛋白采用邻笨三酚红比色法,血浆蛋白采用双缩脲比色法,游离Hb采用Trinder比色法,亚甲蓝采用萃取比色法。结果选出5种试剂盒并建立了室内质控范围;甘油、上清蛋白、血浆蛋白、游离Hb以及亚甲蓝2种仪器比对检测结果均相近(P>0.05);甘油、上清蛋白、血浆蛋白、游离Hb以及亚甲蓝用分光度计各检测48人份,其最高值分别为2.12 g/L、2.40 g/L、63.8 g/L、1.09 g/L、0.31μmol/L。结论血液质量控制项目可采用试剂盒可在分光光度计上进行检测,简单可行,建立室内质控,确保结果准确性。Objective To evaluate test kits performance for glycerin,albumin and plasma protein,free Hb and the feasibility of the methylene blue tests and establish the indoor quality control.Methods Bio reagent kits were purchased directly from the market and were applied to self-made reference products.Thus,a quality control,range can be established.A spectrophotometer and a biochemical analyzer were adopted for parallel tests of glycerine,albumin,plasma protein,free Hb and methylene blue,which are performed by GPO-PAP enzyme colorimetry,phenol red colorimetric method,biuret colorimetry,Trinder colorimetric method,extraction colorimetry,respectively.Results Five kits were picked based on test restults and established correspondent quality control ranges; The two instruments presented no significant difference in terms of measurement for glycerin,albumin and plasma protein,free Hb and methylene blue(P〉0.05); 48 samples were tested for each type and the peak values were 2.12 g/L,2.40 g/L,63.8 g/L,1.09 g/L,0.31 μmol/L,respectively.Conclusion Blood quality control can be performed based on test reagent kits and a spectrophotometer.However,a quality control range need to be deemed to ensure test accuracy.
