摘 要:目的:探讨仙灵骨葆联合康复运动对大鼠骨关节炎(OA)炎性因子mRNA表达的影响。方法:采用前交叉韧带切断术制作大鼠骨关节炎模型。50只SD大鼠随机分为正常对照组、模型组、仙灵骨葆组、康复运动组和仙灵骨葆联合康复运动组。经干预治疗8周后测定大鼠膝关节炎症因子白细胞介素-1β(IL-1β)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)和人基质金属蛋白酶-13(MMP-13)的mRNA表达。结果:与正常对照组比较,模型组、仙灵骨葆组、康复运动组和仙灵骨葆联合康复运动组大鼠的IL-1β、Caspase-3、MMP-13的mRNA表达明显升高,差异均有统计学意义(P<0.05)。与模型组比较,仙灵骨葆组和仙灵骨葆联合康复运动组大鼠的IL-1β、Caspase-3、MMP-13的mRNA表达明显降低,康复运动组大鼠的Caspase-3、MMP-13的mRNA表达明显降低,差异均有统计学意义(P<0.05)。仙灵骨葆联合康复运动组大鼠的Caspase-3 mRNA表达明显低于仙灵骨葆组和康复运动组大鼠,差异均有统计学意义(P<0.05)。结论:仙灵骨葆联合康复运动可以调节骨关节炎诱导的关节退行性改变,减少促炎介质的表达。Adjective:To discuss the effect of Xianling Gubao capsule combined with rehabilitative exercise on mRNA expression of inflammatory factor in osteoarthritis (OA). Methods:Established OA rat models with anterior cruciate ligament transection. Fifty SD rats were divided into the normal control group, model group, Xianling Gubao group, rehabilitative exercise group, Xianling Gubao combined with rehabilitative exercise group. After eight weeks of interventional treatment, detected mRNA expression of interleukin-ll5 (IL-115), caspase-3 and matrix metalloproteinase-13 (MMP-13). Results= Compared with the normal control group, mRNA expression of IL-16, Caspase-3 and MMP-13 of the model group, Xianling Gubao group, rehabilitative exercise group, Xianling Gubao combined with rehabilitative exercise group was increased significantly, differences being significant (P〈 0.05). Compared with the model group, mRNA expression of IL-115, caspase-3 and MMP-13 of the Xianling Gubao group, Xianling Gubao combined with rehabilitative exercise group was decreased significantly, and caspase-3 and MMP-13 of the rehabilitative exercise group was decreased significantly, differences being significant (P〈 0.05). The mRNA expression of Caspase-3 of the Xianling Gubao combined with rehabilitative exercise group was obviously lower than that of the Xianling Gubao group and the rehabilitative exercise group, differences being significant(P〈 0.05). Conclusion= Xianling Gubao capsule combined with rehabilitative exercise can regulate degeneration induced by osteoarthritis and reduce expression of proinflammatory mediators.
