摘  要:目的:了解孕鼠胎盘组织在妊娠期糖尿病(GDM)内环境下脂肪酸转运相关蛋白的水平,探讨维生素E对GDM孕鼠胎盘长链不饱和脂肪酸转运相关蛋白水平的影响。方法:共36只交配成功的C57BL/6J母鼠被纳入研究,其中12只为空白对照(Control组),24只建立GDM孕鼠模型,建模成功后再随机分为GDM组和维生素E补充组(Vit E组)。Vit E组按5mg/(kg·d)维生素E给药,GDM组和Control组灌胃等剂量的溶剂。交配成功后第18天处死孕鼠,检测孕鼠血液中SOD、GSH-px和MAD水平,检测胎盘组织中PPARγ、FATP-1、FATP-4、FAT/CD36和FABPpm蛋白及mRNA水平。结果:GDM组孕鼠的血清SOD、GSH-px和MAD水平显著高于Control组和Vit E组(P<0.05),后两组比较无差异(P>0.05)。GDM组的PPARγ、FATP-1、FATP-4水平低于Control组,FAT/CD36和FABPpm高于Control组,差异均有有统计学意义(P<0.05)。Vit E组的PPARγ、FATP-1、FATP-4较GDM组显著增高(P<0.05),FAT/CD36和FABPpm较GDM组明显降低(P<0.05)。结论:维生素E可降低GDM孕鼠氧化应激水平,上调其胎盘组织PPARγ、FATP-1和FATP-4的水平,下调FAT/CD36和FABPpm水平。Objective: To examine the levels of long chain polyunsaturated fatty acid(LC-PUFA) transport-related proteins and to explore the role of Vitamin E on them in placenta of pregnant rats with gestational diabetes mellitus(GDM).Method: A total of 36 C57 BL/6 J maternal mice with mating were divided into 3 groups,namely Control group,GDM group and Vitamin E supplement group. We used 24 rats of them to establish a model of gestational diabetes mellitus(GDM),then which were averaged into two groups and fed with Vitamin E supplement in one group.Vit E group was treated with 5 mg/(kg·d) Vitamin E,while GDM and Control group were fed with equal dose of solvent.On the 18 th day after pregnancy,the levels of SOD,GSH-px and MAD in the blood were measured,and the levels of PPARγ,FATP-1,FATP-4,FAT/CD36,FABPpm and their mRNA also were detected. Results: The levels of serum SOD,GSH-px and MAD in GDM group were significantly higher than those in control group and Vit E group(P〈0.05).The difference between Control group and Vit E group was not significant(P〉0.05).The expressions of PPARγ,FATP-1 and FATP-4 in GDM group were lower than those in control group(P〈0.05),this trend was reversed in terms of FAT/CD36 and FABPpm(P 〈0.05).Compared with GDM group,the levels of PPARγ,FATP-1 and FATP-4 were significantlyhigher(P〈0.05),and the expression of FAT/CD36 and FABPpm were significantly lower(P〈0.05) in Vit E group.Conclusion: Vitamin E could reduce oxidative stress in GDM pregnant rats by upregulating the levels of PPARγ,FATP-1 and FATP-4 and down-regulating the levels of FAT/CD36 and FABPpm in placental tissue.
