摘  要:【目的】研究建立目的虫种人工感染的单一模式.【方法】采用一种简化的感染性幼虫(L3)鉴别技术,通过测量尾鞘的长度和尾鞘末端的尾丝,从自然混合感染有蛇形毛圆线虫的绵羊粪便中培养并分离该种线虫的L3,然后用10 000条分离的L3经口灌服3~4月龄绵羊.【结果】绵羊感染后第17天从粪便中排出虫卵,第79天虫卵排出量达高峰,此后维持在一个稳定的水平;从人工感染绵羊的粪便中培养和分离出的L3,经鉴定符合蛇形毛圆线虫L3的特征;人工感染10个月后将绵羊剖杀,在其小肠收集到蛇形毛圆线虫成虫2 780条,回收率为27.8%.【结论】成功地建立了蛇形毛圆线虫单种分离株,为今后深入研究寄生线虫的抗药性、生物防治和功能基因等方面奠定了基础.【Objective】The study was to establish monospecific isolate of Trichostrongylus colubriforms.【Method】Using a simplified technique to identify infective third-stage larvae,by measuring the length of the sheath tail and filament at the end of the tail sheath,L3 larvae of T.colubriforms were cultured and isolated from the feces of sheep infected with natural mixed nematodes,and then 3~4 month-old lamb was administered orally with 10 000 L3 larvae isolated.【Result】The results showed that the infected sheep excreted eggs in the feces at 17 th day after infection.The amount of eggs excreted reached the peak on the 79 th day,then it maintained at the stable level.The L3 larvae that were cultured and isolated from feces of sheep artificially infected matched the characteristics of L3 larvae of T.colubriformis by morphological identification.After 10 months in postinfection the sheep was sacrificed and 2 780 adult worms of T.colubriforms were collected in its small intestine and the recovery rate of the larvae to adult stage was27.8%.【Conclusion】A monospecific isolate of T.colubriforms was established successfully and it would lay a foundation for research of anthelmintic resistance,biological-control and functional genes in parasitic nematodes.
