The research of micro-polluted river water purification by in-situ biofilm technology was carried out with sponge iron and ceramsite as the filler. Two kinds of fillers on the treatment effect of (NH4^+-N), total nitrogen (TN), total phosphorus (TP) and chemical oxygen demand (COD) under the conditions of saturated dissolved oxygen (DO) and low DO, as well as sponge iron under reduced hydraulic retention time (HRT) at low DO conditions and ceramsite under the absence of aeration conditions, and the preliminary mechanism analysis and the screening of the filler were discussed. The results showed that, increasing aeration could provide the saturation of DO, but it could impact the biofilm and biomass on the carrier surface. The ceramsite and sponge iron had formed film successfully during 23 days and 56 days respectively, ceramsite was faster than sponge iron. In order to prevent the compaction of sponge, sponge iron could load to mesh polyester ball of 9 cm diameter with 40% volume ratio. Sponge iron in the continuous operation of the removal effect was overall better than ceramsite, and it was more suitable to improve the river water quality under the low DO conditions.
