摘  要:目的探讨氯胺酮单次注射小鼠海马区单羧酸转运蛋白MCT1、MCT2和MCT4的表达变化。方法3月龄C57BL/6小鼠40只随机分为saline组及ket 1组、ket 2组、ket 3组、ket 4组,每组8只,分别腹腔注射生理盐水及10、20、40、80mg/kg氯胺酮,采用Western blot检测不同剂量氯胺酮单次注射后小鼠海马区MCT1、MCT2和MCT4的蛋白表达情况。结果 Western blot结果显示MCT1蛋白在各ket组表达水平均下降(P<0.05),MCT2蛋白仅在ket 3组及ket 4组表达水平升高(P<0.01),MCT4蛋白在各ket组表达水平均下降(P<0.01)。结论单次注射氯胺酮致小鼠脑海马区MCT1和MCT4表达减弱、MCT2表达增强,提示氯胺酮所致神经毒性作用可能与能量代谢受到影响相关。Objective To investigate the changes in the levels of hippocampal MCT1,MCT2 and MCT4 in the mice after single injection of ketamine.Methods Forty C57BL/6 mice were randomly divided into five groups with 8 mice per group treated with the i.p.injections of saline,10 mg/kg ketamine,20 mg/kg ketamine,40mg/kg and 80 mg/kg ketamine respectively.Then western blot were applied to observe the protein levels of MCT1,MCT2 and MCT4.Results Western blot results revealed that the levels of MCT1 and MCT4 significantly decreased compared to saline group(P<0.05),but levels of MCT2 protein exhibited the obvious increase in ket3 and ket4 group(P<0.01).Conclusion Ketamine decreased the hippocampal levels of MCT1 and MCT4 after single administration,whereas the expression of MCT2 were increased.Our results suggest that the disturbance of energy metabolism in brain may be involved in the neurotoxicity of ketamine.
