元江野生稻柱头特异性启动子p51408的克隆及功能分析

摘　　要：【目的】水稻柱头是水稻有性生殖的重要器官,柱头外露是决定异交结实的重要特性,是影响杂交水稻制种和不育亲本繁育的关键因素之一,开展水稻柱头外露特异表达启动子研究是探寻水稻杂种优势分子机理及进行分子育种的基础。【方法】对一个元江野生稻柱头外露特异表达基因的启动子p51048进行了克隆,分析了该启动子的序列结构特征及os51408的表达特性,通过构建该启动子的GUS融合表达载体,获得了转化体。【结果】组化染色分析表明,该启动子能在水稻花器官的柱头、花柱等区域特异表达,而在根、茎叶、颖花中无表达或低表达。【结论】推测其为柱头组织特异性启动子,将为进一步开展水稻柱头外露这一重要性状的分子机理和表达调控研究奠定基础。【Objective 】The stigma exsertion of rice （ saiba L. ） was an important trait determining the outcrossing rates, which was alsoa key factor influencing the hybrid seed production and sterile line breeding, so the research of tissue-specific expression of stigma exsertion of rice would laid the foundation of the molecular mechanism in heterosis of rice. [Method] In this paper, a specific expression promoter p51408 related with stigma exsertion of wild rice （ Oryza rufipogon Griff. ） from Yuanjiang City was cloned, the structural character of its se-quence and expression feature of os51408 was analyzed.【 Result】 With the construction of GUS expression vector and histochemical staining, the results showed that it was specific expression in stigma and style, no or low expression in root, stem, leaf and spikelet.【 Conclusion】 So it could be a tissue-stigma specific expression promoter, and might play a very important role in rice stigma exsertion research.
