摘 要:目的:探究黄柏炮制后化学成分的改变,制定黄柏的质量标准。方法:(1)研究黄柏盐炙的工艺,观察炒制的时间、确定加盐的数量等影响因素,以盐酸小檗红碱和盐酸小檗碱为指标,优选黄柏盐炙工艺。(2)研究盐炙黄柏前后化学成分的改变,采用薄层色谱法,对黄柏盐炙前后化学成分的改变进行比较。(3)研究黄柏炮制后质量情况,研究指标为盐酸小檗碱,炮制后产生的盐酸小檗红碱以及黄柏中盐酸黄柏碱。结果:(1)最佳黄柏盐炙工艺:最优炮制方式为100g黄柏混合2g盐,进行闷润1hr,在150~160℃下炒制8分钟,药材总量的30%水溶解盐。(2)黄柏盐炙后,对化学成分进行测定有部分盐酸小檗碱转化成为盐酸小檗红碱。(3)黄柏炮制后,按照干燥品计算,盐酸小檗碱含量大于等于3.00%,盐酸黄柏碱大于等于0.38%,盐酸小檗红碱大于等于0.08%。结论:(1)经验证,优选的黄柏盐炙工艺炮制方式可行。(2)黄柏成分在盐炙后,部分盐酸小檗碱发生化学反应转化成盐酸的小檗红碱,盐酸黄柏碱含量也有所增加。(3)对黄柏盐炙品的浸出物、总灰分、水分、杂质测定,结果表明所制定的盐黄柏质量标准可行合理。Objective:To study the change of chemical composition of Cortex Phellodendri and the quality standard of Cortex Phellodendri.Methods:1.Study on the process of Cortex Phellodendri stir frying with salt water.By investigating the frying time,the time of smashing,the amount of salt and the temperature of frying,taking berberine hydrochloride and berberine hydrochloride as the index,the changes of chemical composition before and after salt treatment were studied by thin layer chromatography.The quality standard of Cortex Phellodendri was studied by using caffeine hydrochloride,berberine hydrochloride and caffeine hydrochloride as the research indexes.Results:(1) The optimum treatment method was as follows:1 g,100 g of Phellodendron chinensis mixed with 2 g of salt,1 hour of muffin,fried at 150~160 degrees Celsius for 8 min,and 30% of the total amount of the medicine dissolved in salt.(2) Phellodendron chinensis salt after the determination of the chemical composition of some berberine hydrochloride into a small berberine hydrochloride.(3) Cork after processing,according to dry goods,berberine hydrochloride content was greater than or equal to 3.00%,caffeine hydrochloride was greater than or equal to 0.38%,berberine red alkali was greater than or equal to 0.08%.Conclusion:(1) It is proved that the preferred method of processing berberine is feasible.(2)Phellodendron chinensis composition in the salt after the occurrence of quantitative and qualitative change,caffeine hydrochloride increased,some of the berberine hydrochloride converted to berberine hydrochloride.The results show that the quality standard of Phellodendron amurense is feasible and reasonable.
