大承气汤对Wistar大鼠小肠Cajal间质细胞起搏电流影响分析

摘　　要：目的:探讨大承气汤含药动物血浆对Wistar大鼠小肠Cajal间质细胞起搏电流影响,为大承气汤对胃肠道Cajal间质细胞的离子通道的研究提供参考依据。方法:选取8~14天SPF级的Wistar大鼠共20只,雌雄不限,培养大鼠小肠Cajal间质细胞,并采用免疫细胞化学法对其进行鉴定;取Wistar大鼠20只,体重350±20克,雌雄不限,制备服大承气汤含药动物血浆;采用膜片钳技术记录并分析大乘气汤含药动物血浆对Wistar大鼠小肠Cajal间质细胞起搏电流影响。结果:免疫细胞化学法鉴定小肠Cajal间质细胞培养成功。大乘气汤含药血浆条件下,Wistar大鼠小肠Cajal间质细胞起搏电流为~418±60PA,电流振幅相对对照组增加52.6%,差异有统计学意义(P<0.05);频率为15±0.82次/分钟,相对对照组增加76.4%,差异有统计系意义(P<0.05)。结论:大承气汤含药动物血浆对Wistar大鼠小肠Cajal间质细胞的起搏电流有明显的加强作用,大承气汤可通过加强胃肠道Cajal间质细胞起搏电流,改善MODS胃肠运动功能障碍。Objective： To investigate the effect of Dachengqi Decoction on the pacing current of Cajal interstitial cells insmall intestine of Wistar rats, and to provide reference for the study of Dachengqi Decoction on the study of ion channel of Cajalstromal cells in gastrointestinal tract. Methods ： A total of 20Wistar rats of 8 - 4days SPF were selected and the male and femalewere cultured. The Cajal interstitial cells of small intestine were cultured and identified by immunocytochemistry. Twenty Wistarrats＋_20grams, male and female, the preparation of Dachengqi Tang drug containing animal plasma; using patch clamp tech-nique to record and analyze the plasma of Dachengqi Tang plasma Wistar rats small intestine Cajal interstitial cell pacing current.Results ： The culture of small cell Cajal interstitial cells was confirmed by immunocytochemistry. The pulse current of Cajal inter-stitial cells in the small intestine of Wistar rats was -418~60PA, the amplitude of the current was 52. 6% higher than that of thecontrol group, the difference was statistically significant （P〈O. 05） 0. 82times / min, compared with the control group increasedby 76. 4%, the difference was statistically significant （ P〈0. 05 ）. Conclusion： Dachengqi Decoction can enhance the pacemakercurrent of Cajal interstitial cells in the small intestine of Wistar rats. Dachengqi Decoction can improve the gastrointestinal tractCajal interstitial cell pacing current and improve the MODS stomach Intestinal motility dysfunction.
