摘 要:目的介绍一种经济实用、成功率高的气管插管方法来制备矽肺大鼠模型。方法将20只SPF级Wistar雄性大鼠随机分为对照组和矽肺模型组,2组均使用自制实验器具通过直视下行插管法给药,经检验插管成功后向对照组气管注入1 ml生理盐水,模型组气管注入1 ml Si O2悬浊液。染毒后28 d取出肺组织行HE和Masson染色,应用碱水解法检测肺组织中羟脯氨酸含量。结果 2组大鼠造模过程中无死亡;造模后对照组大鼠生长状况良好,模型组大鼠生长缓慢并伴有毛枯、喘息等症状;肺组织病理学检查发现,对照组大鼠肺组织结构正常,无明显纤维化;模型组大鼠肺组织出现典型的肺泡炎和纤维化改变。模型组大鼠羟脯氨酸含量明显高于对照组,差异有统计学意义(P<0.01)。结论组织病理学检查及羟脯氨酸测定结果提示经改良的气管插管注药方法成功复制了矽肺模型,操作简便,死亡率低,可加以推广。Objective To introduce an economical and practical method of silicosis rat model by tracheal intubation. Methods A total of wistar male rats of SPF grade were randomly divided into control group and silicosis model group. The two groups were treated with an improved intubation method to ferry drugs into lung under direct vision using self-made experimental equipment. After successfully inspect intubation, the control rats were injected with normal saline solution (lml per rat), and in the model rats were injected with silicon dioxide (lml per rat). The HE and Masson staining of rat lung tissue was performed and the hydroxyproline contents were measured by alkali hydrolysis method after 28 days. Results There was no death during the molding process. After molding, the rats in control group were in good condition, and the symptoms of the rats in the model group was slow-growing, and accompanied by the symptoms such as the dry hair and polypnea. The histopathological examination of lung tissue showed that the structure of rat lung tissue in the control group was normal, and there was no obvious fibrosis, while the model group showed typical pulmonary alveolitis and fibrosis. The contents of hydroxyproline in the model group were significantly increased compared with the control group (P〈0.01). Conclusion The results of the histopathological examination and hydroxyproline contents point out that the silicosis rat model was successfully established by the modified endotraeheal intubation. The method had the advantages of easy operation and low mortality, and is worth to be popularized.
