摘  要:目的探讨外周血辅助型T细胞分泌的细胞因子在特发性血小板减少性紫癜(ITP)模型小鼠的变化及意义;方法采用CBA方法分别检测14只正常小鼠,14只未治疗ITP模型小鼠及14只泼尼松治疗后ITP模型小鼠外周血血清中的IL-2、INF-γ(Th1)、IL-4、IL-10(Th2)、TGF-β1(Th3)和IL-17(Th17)细胞因子水平的变化;结果与正常组比较,模型组中的IFN-γ、IL-2(Th1)、IL-17(Th17)水平明显升高,IL-10、IL-4(Th2)、TGF-β1(Th3)水平明显降低,Th1/Th2比值增高(P均<0.05),泼尼松组与正常组比较,Th1、Th2、Th17和Th3型细胞因子水平及Th1/Th2比值无明显差异(P均>0.05);结论在ITP模型小鼠中存在细胞亚群平衡的偏移,表现为Th1、Th17型细胞相关细胞因子分泌亢进,Th2、Th3型细胞相关细胞因子分泌减低,提示辅助型T细胞相关细胞因子参与了ITP的发病机制。Objective To investigate the changes and significance of idiopathic thrombocytopenic purpura (ITP)model mice peripheral blood T helper cell secreted in cytokines;Methods Used the method of Ctyometric Beads Arrays to detect cytokines levels of changes of IL- 2 and INF-γ( Th1), IL- 4 and IL- 10 (Th2) ,TGF-β1 (Th3),IL-17 (Th17)of 14 normal mice, 14 mice with untreated ITP and 14 treated with prednisone ITP mouse model ; Results Compared with the normal group, the leves of IFN-γ and IL-2 ( Th1 ), IL- 17 ( Th17 ) ,the ratio of Th1/Th2 were elevated ; the level of IL- 10 and IL-4 ( Th2 ) ,TGF- β1 ( Th3 ) were desreased in model group(P 〈0.05 ). Compared with the normal group,the levels of Th1,Th2,Th3 and Th17 type cell factors were not significantly different in prednisone group (P 〉 0.05 ) ;Conclusion The existence of cell subsets balance shift in ITP mouse model; Performance of Th1 and Th17 cytokines secretion hyperthyroidism, Th2 and Th3 cytokines secretion reduction, pointed to T helper cell eytokines involved in the pathogenesis of ITP.
