摘 要:目的观察γ-干扰素(IFN-γ)刺激、活化的骨髓来源巨噬细胞(BMDMs)对小鼠神经干细胞(NSCs)迁移的影响。方法分离、原代培养骨髓巨噬细胞和NSCs,然后通过体外迁移实验比较对照组、巨噬细胞组和巨噬细胞+IFN-γ组迁移的NSCs数量。同时,利用酶联免疫吸附实验(ELISA)检测IFN-γ刺激后巨噬细胞单核细胞趋化蛋白-1(MCP-1)的含量。结果成功体外培养高表达F4/80的巨噬细胞和高表达巢蛋白(Nestin)、具有多向分化潜能的NSCs。迁移实验结果显示,与对照组相比,巨噬细胞组和巨噬细胞+IFN-γ组迁移的NSCs数量显著增加(P值分别为0.007、0.000)。巨噬细胞+IFN-γ组[(0.75±0.05) mm2]较巨噬细胞组[(0.25±0.03) mm2]迁移的NSCs数量增加,差异有统计学意义(P=0.023)。ELISA结果显示巨噬细胞+IFN-γ组[(650±65) pg/ml]较巨噬细胞组[(420±23) pg/ml]上清液中MCP-1含量显著升高(P=0.037)。结论IFN-γ刺激巨噬细胞分泌可溶性因子诱导NSCs迁移,其中MCP-1可能起重要作用。Objective To investigate the effect of bone marrow derived macrophages on the migra- tion of neural stem cells (NSCs) under the stimulation of interferon - γ(IFN -γ). Methods Bone marrow derived macrophages and NSCs were isolated and cuhured. The migration of NSCs in different groups were detected by the migration chamber. At the same time, the levels of monocyte chemotactic protein - 1 ( MCP - 1 ) were tested using the enzyme - linked immunosorbent assay (ELISA) after macrophages in- duced by IFN -γ,. Results NSCs which expressed highly nestin and had the ability to differentiate into neuron and glial cells, and F4/80 positive macrophages were successfully cultured in vitro. Compared with the control group, the numbers of migrated NSCs increased significantly in macrophages group and macro- phages + IFN - γ group (P = 0. 007,0. 000 ). While macrophages remarkably increased the migratory abili- ty of NSCs under the stimulation of IFN -γ [ (0. 75±0. 05) mm^2 vs. (0. 25±0. 03) mm^2, P =0. 0231. ELISA results showed that the level of MCP - 1 macrophages + IFN -γ group increased, compared with macrophages group (P = 0. 037 ). Conclusion Macrophages enhance the migration on NSCs under the stimulated of IFN -γ and MCP - 1 may play an important role on it.
