摘 要:目的观察高纯度蛇毒因子(CVF)等多因素处理对异位辅助肝移植术后免疫功能的影响。方法建立猪→猴异位辅助肝脏移植模型,观察高纯度CVF等多因素处理后,异位辅助肝移植超急性排斥反应(HAR)和延迟性异种排斥反应(DXR)发生的状况,比较免疫吸附后的移植肝组织和受体死亡时移植肝组织内免疫球蛋白G(IgG)、免疫球蛋白M(IgM)及补体C3蛋白的沉积情况,了解CVF及免疫吸附等多因素处理对猪→猴异种肝移植后免疫功能的影响。结果用于吸附的供体肝脏均发生HAR,HAR发生的时间分别为10、12、11、9、13 min,平均(11.00±1.41) min;病理表现为镜下见组织内缺血性坏死和血栓形成。经CVF等多因素处理后,受体猴的移植肝均存活到受体死亡时,所有受体猴移植肝经过病理学检查均未发生HAR和DXR,受体死亡后移植肝组织内IgG、IgM和C3蛋白的沉积均低于免疫吸附后的供体肝组织,差异有统计学意义(P=0.026、0.047、0.032)。结论CVF等多因素处理能有效克服猪→猴异种移植HAR的发生,并有可能阻止或延缓DXR的发生。Objective To explore the eft-ct of high purity venom factor (CVF) and other factors on immune function after pig to monkey liver xeno - transplantation. Methods To establish the pig to monkey heterotopic auxiliary liver transplantation model, the occurrence of super acute rejection (HAR) and delayed rejection (DXR) was observed after liver transplantation while interfered by high purity CVF. The expression of IgG, IgM and C3 protein in liver tissues was detected by Western blotting at different time points. Results The donor liver used for the adsorption all occured HAR, respectively of 10, 12, 11, 9, 13 minutes, the average time was (11.00 ± 1.41 ) minutes. The pathological findings were ische-mie necrosis and thrombosis. After CVF and other multiple factors, all the recipient monkey liver did not observe HAR and DXR until death, the IgG, IgM and C3 protein deposition in liver tissue after the death of the receptor were lower than the donor liver used for the adsorption, which is statistically significant difference (P = 0.026, 0. 047, 0, 032). Conclusion CVF and other multiple factors can effectively overcome the occurrence of HAR in pig to monkey liver xeno - transplantation, and has the potential to prevent or de- lay the DXR.
