“十伤灵”中药膏对难愈性皮肤溃疡模型大鼠疡面愈合的影响研究

摘　　要：目的:探讨"十伤灵"对难愈性模型大鼠溃疡面愈合的影响。方法:将41只Wistar大鼠随机分为空白组、创灼膏组(CZ组)、十伤灵组(SS组)、模型组。其中36只大鼠背部制造难愈性溃疡,分别将药物涂抹在各组大鼠背部创面处。于造模后7 d、14 d分别取材通过Nagelschmidt法观察对比各组创面愈合率和愈合时间;采取免疫组织化学染色及定量分析,比较各组皮肤创面bFGF及其受体bFGFR的表达量;利用显微镜图像分析仪结合Nagelschmidt法,测量疤痕组织切片中成纤维细胞形成的瘢痕面积。结果:实验第7天、第14天,各组创面愈合率比较,差异均无统计学意义(P>0.05)。实验第14天,模型组、创灼膏组和十伤灵组bFGF值均高于空白组(P<0.05);模型组、创灼膏组和十伤灵组bFGF比较,差异无统计学意义(P>0.05)。实验第7天,模型组、创灼膏组的成纤维细胞数多于空白组(P<0.05),而十伤灵组与空白组比较,差异无统计学意义(P>0.05);模型组、创灼膏组和十伤灵组成纤维细胞数比较,差异无统计学意义(P>0.05)。实验第14天,模型组、创灼膏组的成纤维细胞数比较,差异无统计学意义(P>0.05);十伤灵组成纤维细胞数低于模型组(P<0.05)。结论:"十伤灵"对大鼠背部的难愈性溃疡面有促进修复作用。Objective： To explore the effect of Shishangling（十伤灵） on the healing of chronic skin ulcer rat model of surface ulcers.Methods： The 41 Wistar rats were randomly divided into four groups： blank group,＂Chuangzhuogao＂（创灼膏） group（CZ）,＂Shishangling＂ group（SS）,the control group（5 rats）,36 rats were made manufacturing difficult prognosis ulcer in the back,respectively give drug in each group of rat back wound.After 7 d and 14 d,the wound healing rate and healing time were observed and compared by Nagelschmidt method.Immunohistochemical staining and quantitative analysis was used to compare the expression of bFGF and its receptor bFGFR in skin wounds of each group.The scar area of fibroblasts in scar tissue sections was measured by microscope image analyzer combined with Nagelschmidt method.Results： There was no significant difference in wound healing rate between the groups at 7 thday and 14 thday（P〉 0.05）.On the 14 thday,the bFGF values of model group,Chuangzhuogao group and Shishangling group were higher than that of blank group（P〈 0.05）; there was no significant difference in bFGF between model group,Chuangzhuogao group and Shishangling group（P〉 0.05）.At the 7 thday of the experiment,the model group,fibroblast cells of Chuangzhuogao group was more than the control group（P 〈0.05）,and Shishangling group compared with the control group,the difference was not statistically significant（P 〉0.05）.On the fourteenth day,there was no significant difference in the number of fibroblasts between the model group and the Chuangzhuogao group（P〉0.05）; the number of fibroblasts in the Shishangling group was lower than that in the model group（P〈 0.05）.Conclusion： Shishangling can promote ulcer repair of rat dorsal surface.
