摘 要:目的分析Mylpf基因在糖尿病长爪沙鼠6种组织中的表达水平,进而探究长爪沙鼠糖尿病发生的分子机制。方法选取糖尿病长爪沙鼠和正常沙鼠各6只,使用Real-time PCR和Western blotting技术分别检测动物的骨骼肌、肝脏、脂肪、肾脏、心脏和脑组织中Mylpf的mRNA和蛋白表达水平。结果 Real-time PCR的结果表明,在骨骼肌、心脏和脑组织中,Mylpf在糖尿病组mRNA表达水平高于对照组。在脂肪组织和肝脏组织中糖尿病组mRNA表达水平低于对照组。在肾脏组织中两组mRNA表达水平基本相同。Western blotting结果显示,在骨骼肌中Mylpf在糖尿病组蛋白表达水平高于对照组,且有统计学差异,在心脏和脂肪组织中Mylpf在糖尿病组蛋白表达水平低于对照组,在肾脏组织中蛋白表达水平基本相同,在肝脏和脑组织中没有检测出。结论 Mylpf与长爪沙鼠糖尿病的发生有一定相关性,该基因对长爪沙鼠糖尿病的影响可能主要发生在骨骼肌和心脏组织中。Objective To analysis of Mylpf expression levels in six tissues of diabetic Mongolian gerbils and to explore the molecular mechanism of diabetes in Mongolian gerbils.Method We selected a diabetic group and a control group,each group has 6 Mongolian gerbils.Mylpf mRNA and protein expression level in skeletal muscle,liver,adipose,kidney,heart and brain were separately detected using Real-time PCR and Western blotting.Result The Real-time PCR showed that compared with control animal,mRNA expression of Mylpfin skeletal muscle,heart and brain was increased in diabetic group.The relative expression level in diabetic group exhibited decreased compared with control in adipose and liver.And there were no differences between two groups in the kidney.The Western blotting result display that protein of Mylpf in skeletal muscle was significantly increased in diabetes group.The expression of Mylpf in diabetic Mongolian gerbils heart and adipose was lower than control group.Mylpf protein was almost undetectable in liver and brain tissue.And Varied expression was observed in kidney,but there is no difference.Conclusion There is a correlation between Mylpf and the occurrence of diabetes in Mongolian gerbils and theresult show that the effect of Mylpf mainly occurred in skeletal muscle and heart tissue of diabetic gerbils.
