摘 要:目的研究温郁金挥发油对β-淀粉样蛋白(Aβ_(25-35))致阿尔茨海默病(AD)模型小鼠的学习记忆改善作用。方法 60只昆明种小鼠随机分为假手术组,模型组,温郁金挥发油低(0.016 mL/kg)、高(0.048 mL/kg)剂量组,盐酸多奈哌齐组(1.30 mg/kg)及参枝苓组(2.6 mL/kg),海马内注射(Aβ_(25-35)),制作小鼠AD模型。连续灌胃给药15 d。Y迷宫和Morris水迷宫实验检测各组小鼠认知行为的改变,HE染色观察其对AD模型小鼠神经元的影响。结果温郁金挥发油可显著增加AD模型小鼠的自发交替反应率,缩短逃避潜伏期,减少总路程,延长游泳路程百分比,具有统计学意义(P<0.05或P<0.01);可改善海马区神经元细胞排列紊乱、水肿等现象,减少神经元死亡。结论温郁金挥发油对Aβ_(25-35)致阿尔茨海默病模型小鼠学习记忆能力具有改善作用。Objective To investigate the effects of curcuma aromatica volatile oil on learning and memory in mice induced by β-amyloid( (Aβ25-35).Method Sixty mice were divided into six groups randomly: sham group,model group,curcuma aromatica extract low dose group (0.016 m L/kg),high dose group (0.048 m L/kg),donepezil group (1.30 mg/kg) and shenzhiling group (2.6 m L/kg).β-Amyloid protein (Aβ25-35) was infused into the hippocampal of mice.Drugs were administrated for 15 days.Y maze and Morris water maze was detected to evaluate the cognitive behavior.HE-staining was used to observe the neuron changes caused by curcuma aromatica volatile oil.Result Compared with the model group,curcuma aromatica volatile oil increase the alternation behavior significantly,decrease swimming escape latency and total distance,prolong the swimming distance percentage (P 〈 0.05 or P 〈 0.01); It also can improve the disorder,edema and other symptoms of hippocampus neurons,reduce the death of neurons.Conclusion Curcuma aromatica volatile oil can improve the effects of learning and memory in mice induced by β-amyloid (Aβ25-35).
