摘  要:目的观察和分析TGF-β1对子宫内膜基质细胞(endometrial stromal cells,ESCs)增殖和活性的影响。方法体外分离、培养和鉴定ESCs。实验分组:对照组和TGF-β1组,对照组:加入不含血清的培养液;转化生长因子-β1(TGF-β1)组:不含血清的培养液+不同浓度的TGF-β1,通过噻唑蓝法检测各组细胞的增殖率;用ELISA法检测各组细胞培养液中Ⅰ型胶原(Collagen typeⅠ,ColⅠ)和纤黏连蛋白(Fibronectin,FN)的含量。免疫印迹法检测各组细胞胞质中的MMP-9及TIMP-1的表达水平。结果 TGF-β1可以促进ESCs增殖,并呈剂量依赖性;使细胞分泌ColⅠ(0.736 6±0.042 4)和FN(0.713 1±0.046 8)的量增多,细胞内MMP-9含量减低,而TIMP-1含量升高;与对照组比较,其差异有统计学意义(P<0.05)。结论 TGF-β1能使子宫内膜基质细胞增殖,并使其活性增加。[Objective] To observe and explore the effect of TGF-β1 on proliferation and activity of human endometrial stromal ceils (ESCs). [Methods] The ESCs was isolated,cultured and identified in vitro. The experiment groups include: control group and TGF-β1 groups. Control group: ESCs was cultivated by solution without serous. TGF-β1 groups: ESCs was cultivated by solution (without serous) with different concentration of TGF-β1.The method of MTF was used to detect the proliferation of ESCs. The contents of fibronectin (FN) and collagen type I (Col I ) in the cultured supernatants were measured by enzyme-linked immunosorbent assay (ELISA). Western blot was used to detect the expression levels of MMP-9 and TIMP-1. [ Results] TGF-I31 improved the proliferation of ESCs,with a dose-dependent effect. The contents of Col I (0.736 6±0.042 4) and FN(0.713±0.046 8) increased,MMP-9 level decreased and TIMP-1 level increased,and the differences were statistically significant between control group and TGF-β1 groups (P〈O.OS).[Conclusion] TGF-β1 can promote ESCs proliferation and increase activity of ESCs.
