摘  要:目的:探讨高良姜、胡椒和槟榔有效成分对岗田酸诱导p-tau细胞模型的作用。方法:采用CCK8法确定48 h内岗田酸,高良姜素、胡椒碱和槟榔碱对高分化PC12细胞的作用浓度,随之将其分为空白对照组,模型对照组、高良姜素组、胡椒碱组和槟榔碱组,检测细胞中p-tau含量和观察细胞形态变化。结果:在48 h内,岗田酸的造模适应浓度是175 n M,高良姜素、胡椒碱和槟榔碱的LD50分别是150 pM、24 pM和250 pM。与空白对照组比较,模型对照组的p-tau显著增加(P<0.01),说明岗田酸诱导p-tau细胞模型造模成功;与模型对照组比较,高良姜素组、胡椒碱组和槟榔碱组的p-tau显著减少(P<0.01),细胞形态的损伤减少。结论:高良姜素、胡椒碱和槟榔碱对岗田酸诱导p-tau模型细胞有一定的作用。文献引用:赵贤武,黄丽平,邓敏贞,等.高良姜、胡椒和槟榔有效成分对岗田酸诱导p-tau细胞模型的作用研究[J].中医学报,2017,32(11):2176-2180.Objective: To study the effect of the component of Galangal,Pepper and Areca on p-tau cell model induced by Okadaic Acid( OA). Methods: The concentration of OA,Galangin,Piperine and Arecoline for high differentiated PC12 cell in 48 hours was detected by CCK8 method. Then it can be divided into blank control group,model control group,Galangin treated-group,Piperine treated-group and Arecoline treated-group. P-tau content and morphological changes were tested. Results: The LD50 of OA,Galangin,Piperine and Arecoline within 48 h was 175 n M,150 pM,24 pM,and 250 pM. P-tau in the model control group increased significantly compared with that in the blank control group( P 0. 01). It meaned that p-tau cell model induced by OA was successfully completed. P-tau levels of Galangin treated-group,Piperine treated-group and Arecoline treated-group were significantly decreased( P 0. 01) and cell morphology damage reduced compared with those in the model control group. Conclusion: Galangin,Piperine and Arecoline have effect on p-tau cell model induced by OA.
