摘  要:利用响应面法系统优化复合诱变纳豆芽孢杆菌N-E5.2-RE生产γ-聚谷氨酸的发酵培养基成分.通过单因素试验、Plackett-Burman试验、最陡爬坡试验及Box-Behnken试验构建响应方程,可预测得到最优培养基(g·L^(-1)):细菌学蛋白胨15、蔗糖46.375、谷氨酸钠45.85、酵母提取粉2.5、NaCl 5、MgSO_40.5、MnSO_4·H_2O 0.4、CaCl_20.25、K_2HPO_42和KH_2PO_45.1.此条件下,pH调至7.3-7.4后,将2%菌株N-E6-RE接种到200mL的锥形瓶中,37℃、200r·min^(-1)摇瓶发酵48h,γ-PGA产量达到38.94g·L^(-1),底物谷氨酸钠转化率比优化前提高了70.8%.Response surface methodology is used to optimize fermentation medium components produced by mutagenic Bacillus natto N-E5.2-RE of poly-γ-glutamic acid.By single factor experiment,Plackett-Burman experiment,steepest ascent test and Box-Behnken experiment building response equation,the optimal medium(g·L^-1)is predicted:peptone 15,sucrose 46.375,sodium glutamate 45.85,yeast extract powder 2.5,NaCl 5,MgSO40.5,MnSO4·H2 O 0.4,CaCl20.25,K2 HPO42 and KH2 PO45.1.Under these conditions,pH is adjusted to 7.3-7.4 and the 2% Bacillus natto NE6-RE is inoculated into Erlenmeyer flask whose capacity is 200 mL,so the production ofγ-PGA could reach 38.94 g-L^-1 at 37℃ with shaking at a speed of 200 r·min^-1 for 48 h,and the conversion rate of the substrate glutamate sodium glutamate is 70.8% higher than before the optimization.
