摘 要:目的:建立一种能延长薄荷种质资源离体保存时间的方法。方法:以薄荷试管苗为材料,接种到添加不同配比的甘露醇和蔗糖的MS(Murashige and skoog,1962)培养基中;在温度为(25±1)℃、光照强度2000 Lx,光照时间12 h/d、相对湿度60%左右的环境下培养;12个月后选成活率较高的试管苗转接到生根培养基进行恢复生长后驯化盆栽,以正常继代苗作对照,分别测量其株高、茎粗、叶片数及叶片SPAD值。结果:试管苗在添加蔗糖浓度30~90 g/L和甘露醇浓度0~15 g/L的MS培养基短期内都有部分存活,最长能保存12个月,存活率最高达60%。保存12个月的试管苗在MS培养基上均能恢复正常生长,驯化盆栽后的成活率为100%。结论:添加浓度为70 g/L蔗糖和5~10 g/L甘露醇的MS培养基适合薄荷试管苗的离体保存。Objective:To establish a method for extending the conservation time of mint resources in vitro. Meth, otis:The tubes of mint were inoculated into MS (Murashige and skoog, 1962 ) medium with different ratios of mannitol and sucrose, and cultured under the condition that the temperature was (25 ± 1 ) ℃, the illumination was about 2000 Lx, 12 h / d and the relative humidity was about 60 % . After 12 months, the two groups with higher survival rates were transferred to the rooting medium to restore the growth and then cultured in pots; and the normal subcultures were used as the control. The plant height, stem diameter, leaf number and relative Chlo- rophyll SPAD were measured. Results:The tube plants of all groups could partially survive when the sucrose and mannitol concentrations were 30 - 90 g / L and 0 - 15 g / L respectively in a short period of time , and the high- est survival rate was 60 % after being cultured for a year. After recovered growth and potted planting, the per- centage of survival could reach to 100 %. Conclusion:The MS medium with 70 g / L sucrose and 5 - 10 g / L mannitol was suitable for conservation of the tube plant of mint.
