摘 要:猪传染性胃肠炎病毒(TGEV)TH-98株是1998年在黑龙江省哈尔滨市郊区疑似感染TGEV的猪场中分离出来的高致病性毒株。采用RT-PCR以及RACE技术扩增出TGEV的全基因组序列并上传至Gen Bank,将TH-98株同Gen Bank中其他13株TGEV进行比较。结果表明14株TGEV Nsp1~Nsp16、S、ORF3a、ORF3b、E、M、N和ORF7氨基酸序列同源性分别为98.0%~100%、97.3%~99.8%、91.2%~100%、27.0%~100%、93.8%~100%、99.6%~100%、97.3%~100%和92.3%~100%。与已发表的13株TGEV毒株相比,TH-98株Nsp1~Nsp16、E、N及ORF7基因均无缺失或插入;与Miller M60的M基因相比,TH-98株有6nt的缺失。不同的TGEV株的突变、缺失、插入主要发生在ORF3b和S基因中。进化树分析可知TGEV TH-98与Purdue株亲缘关系较近,同Miller株拥有共同的祖先。TH-98株同近几年分离出的毒株JS2012、TGEV-HX、SHXB全基因序列同源性分别为98.6%、98.8%、99.8%。研究表明,TH-98株与目前流行的TGEV毒株相比没有发生明显的变化。In 1998,a highly pathogenic strain of transmissible gastroenteritis virus(TGEV),TH-98,was isolated from one pig farm with suspected TGEV in the Harbin suburb of Heilongjiang Province.The complete genome of strain TH-98 was obtained by RT-PCR and RACE technology and uploaded to the NCBI,and then its sequence was compared with those of other thirteen TGEV strains in the Gen Bank database.The results showed that the homology of the amino acid sequences of the nonstructural proteins Nsp1 to Nsp16,S,ORF3 a,ORF3 b,E,M,N,ORF7 between strain TH-98 and other TGEV strains was 98.0% to 100%,97.3% to 99.8%,91.2% to 100%,27.0% to 100%,93.8% to 100%,99.6% to 100%,97.3% to 100% and 92.3% to 100%,respectively.And there were no insertions or deletions in the nonstructural genes ORF1 and ORF7,or in the genes encoding the structural proteins such as envelope(E) and nucleoprotein(N).Compared with membrane(M) gene of Miller M60,there was 6 nt deletion in that of TH-98 strains.The common phenomenon in all TGEV strains was that the mutation,insertion and deletion was mainly observed in ORF3 b and spike(S) gene.Phylogenetic analysis showed that strain TH-98 was closely related to the Purdue strains and shared a common ancestor with the Miller strains.Compared with JS2012,TGEVHX and SHXB strains isolated in recent years,the homology of the complete genome was 98.6%,98.8% and 99.8%,respectively.It suggests that there are no obvious changes between strain TH-98 and the current epidemic strains.
