摘 要:目的:探究黄秋葵水提液对糖尿病小鼠的治疗作用及作用机制.方法:用四氧嘧啶进行糖尿病小鼠造模;用黄秋葵水提物对糖尿病小鼠进行灌胃,考察糖尿病小鼠的血糖变化.利用分子对接方法,考察黄秋葵所含成分对α-葡萄糖苷酶的抑制作用.结果:黄秋葵水提液能够降低糖尿病小鼠的血糖(P≤0.01),并呈一定剂量依赖性.黄秋葵中所含甾醇类成分可与α-葡萄糖苷酶稳定结合,其中有5个化合物与α-葡萄糖苷酶结合自由能在-9.9 kcal/mol以下.结论:黄秋葵水提液能够有效降低糖尿病小鼠的血糖,其作用机制可能是其中所含的甾醇类成分抑制α-葡萄糖苷酶的活性.AIM: To investigate the effect of water extract of oksa on diabetes mellitus( DM) model mice and the underlying mechanism. METHODS: The diabetic mice model was established by feeding alloxan; The diabetic mice was fed with water extract of oksa once a day; Then the variations in the blood glucose of the model mice was measured daily. The inhibitors ofα-glucosidase contained in oksa was screened with molecular docking method. RESULTS: Water extract of oksa can reduce the concentration of blood glucose of model mice( P≤0.01) in a dose-dependant way. Sterol ingredients contained in oksa can stabilize with α-glucocidase,among which there are five compounds with binding free energies lower than-9. 9 kcal/mol.CONCLUSION: Oksa may play a role in reducing the blood glucose of model mice in which α-glucocidase activity was inhibited by its sterol ingredients.
