摘  要:研究应用RNA-seq和聚合酶链式反应确定了一个热休克同源蛋白70-2(MsHSC70-2)基因以及应用荧光定量PCR检测紫花苜蓿不同季节和不同器官中MsHSC70-2的mRNA含量。结果表明:该基因的mRNA序列含有2 025bp,开放阅读框1 950bp,编码649个氨基酸,该基因的蛋白分子量约70.97kDa,等电点4.74。同一器官不同季节MsHSC70-2mRNA含量不同(P<0.05),夏季时不同器官间含量存在差异(P<0.01),该基因在根、茎、叶和顶芽中的表达趋势从四月份到九月份都是先升高后降低,表明MsHSC70-2的表达受日照长度和温度的调节,其表达量变化影响了各器官的生长发育,高表达促进苜蓿的生长发育,低表达抑制苜蓿的生长发育,该基因表达的降低可能参与秋眠型苜蓿秋眠的发生。研究结果为进一步研究紫花苜蓿HSC70-2的功能提供了依据。In this study, a MsHSC70-2 gene was determined by polymerase chain reaction (PCR), and its mRNA sequence is 2025bp, which contains a complete open reading box of 1950bp, encoding 649 amino acids, molecular weight of the protein is about 70.97 Kda, its isoelectric point is about 4.74. The mRNA content of MsHSC70-2 from different organs of the variety in different seasons were detected by qRT-PCR, showing that its content in the same organs had significant difference in different seasons(P〈0.05), and the content of MsHSC70-2 mRNA among organs was different during summer(P〈0.01). The expression trend of the gene in root, stem, leaf and bud increased first and then decreased from April to September,demonstrating that the expression of MsHSC70-2 is regulated by day length and temperature, and its expression variation affects the growth and development of various organs, its high expression promotes the growth and development of alfalfa, and plant growth was inhibited at low expression level,showing that decrease of its expression may participate in fall dormancy in alfalfa.The study provides significant basis for further study on the function of alfalfa HSC70-2.
