摘 要:目的:五指毛桃为广东地区习用药材,商品中存在较多混淆品。该文拟对五指毛桃及其可能的混淆品进行鉴别研究。方法:采用TLC法对五指毛桃及其混淆品进行定性鉴别,同时采用HPLC法进行定量分析,以及利用ITS2序列进行DNA条形码鉴定。结果:TLC鉴别检出五指毛桃和琴叶榕中均含有补骨脂素,而变叶榕、对叶榕和构树中则未检出补骨脂素;建立了基于HPLC法检测五指毛桃及其混淆品中补骨脂素、佛手柑内酯与芹菜素的定量方法;建立了基于ITS2序列的五指毛桃及其混淆品的DNA条形码,NJ树显示五指毛桃及其混淆品可从分子水平上得以区分。结论:综合运用不同的鉴定方法可对五指毛桃及其混淆品进行准确和全面的鉴定,为五指毛桃的鉴别提供新的科学依据。Objective:To study the authentication methods of the Fici Hirtae Radix which had been one of the most extensively used medicines in Guangdong province. Methods:TLC method was applied to analyze the qualitative identification of Fiei Hirtae Radix and its adulterants. HPLC method was applied to analyze the content of Fici Hirtae Radix and its adulterants at the same time. Fici Hirtae Radix and its adulterants were identified by ITS2 sequences of DNA barcoding. Results :TLC showed that Fici Hirtae Radix and Fici Panduratae Radix contained psoralen, but others were not checked it out. Further, to establish a HPLC method for determining the con- tents of psoralen, bergapten and apigenin. DNA bacording establishment was based on ITS2 sequence in order to identify Fiei hirtae radix and its adulterants, NJ tree showed that they could be distinguished from molecular level obviously. Conclusion : The integrated use of identifiable methods for Fici Hirtae Radix and its adulterant is accurate and comprehensive appraisal, which provides new scientific ba- sis for the identification of Fici Hirtae Radix.
