摘 要:目的:探讨木豆叶对心肌缺血-再灌注损伤大鼠心功能的影响及其可能的作用机制。方法:60只健康成年SD大鼠被随机分成6组,每组10只。结扎大鼠左冠状动脉前降支(LAD)30 min,再灌注90 min,在不同时间点记录大鼠心率(HR)、平均动脉血压(MAP)、左室发展压(LVDP)、左室内压最大上升速率(+dp/dtmax)和左室内压最大下降速率(-dp/dt_(max));测定大鼠血清中丙二醛(MDA)含量及乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GXH-Px)、一氧化氮(NO)和肌酸激酶(CK)的活性。结果:缺血后模型组血流动力学指标开始逐渐下降,除HR外,MAP、LVDP及±dp/dt_(max)较T_0时显著降低(P<0.05);应用木豆叶后,各指标在某些时间点虽仍低于T0时刻,但差异无统计学意义。缺血-再灌注可导致大鼠血清MDA含量和LDH、CK活性显著升高,SOD、NO、GSH-Px活性显著降低(P<0.05),应用木豆叶能显著对抗这种变化。结论:木豆叶能对抗心肌缺血-再灌注引起的大鼠心功能紊乱,作用机制可能与其抗氧化有关。Objective:To investigate the effects and mechanism of Cajanus cajan leaves on functional influence in rat hearts with myocardial ischemia-reperfusion(IR)injury. Methods:60 rats were randomly divided into six groups,with ten rats in each group. IR in- jury was induced in rat hearts by ischemia for 30 min and then reperfused for 90 min. The heart rate ( HR), mean arterial pressure (MAP), the left ventricular developed pressure (LVDP), maximum internal pressure of the left ventricle ( + dp/dt max ), and the drop rate of left ventricular pressure ( - dp/dt max)was measured by using a pressure transducer. The content of malondialdehyde ( MDA), the activity of lactate dehydrogenase ( LDH), superoxide dismutase ( SOD), glutathione peroxidase ( GXH-Px), nitric oxide ( NO ), and crea- tine kinase(CK) in the myocardium were detected using commercial kits. Results :The hemodynamic parameters of model group began to decline after ischemic. MAP, LVDP and + dp/dt max were decreased significantly when compared with of To time( P 〈 0.05 ). Pretreatment with Cajanus cajan leaves before ischemia can inhibite the hemodynamic parameters decreasing(P 〉 0. 05). Myocardial ischemia-reperfusion significantly increased the myocardial content of MDA, and the activities LDH and CK,while decreased the SOD, NO and GSH- Px activities,pretreatment with Cajanus cajan leaves markedly antagonized the effect of isehemia-reperfusion. Conclusion:Cajanus cajan leaves can inhibit ischemia-reperfusion injury which induced cardiac function disorder, the mechanism underlying the effect of Cajanus cajan leaves may be associated with its antioxidant capacity.
