摘 要:目的:建立不同产地野生和市售山楂叶的UPLC指纹图谱。方法:采用UPLC法,色谱柱为ACQUITY UPLC BEH C_(18)柱(100 mm×2.1 mm,1.7μm),以乙腈-0.1%甲酸溶液梯度洗脱,柱温30℃,流速0.3 mL/min,检测波长355 nm,通过采用相似度分析、主成分分析(PCA)以及正交偏最小方差判别分析(OPLS-DA)等化学计量分析方法来评估药材的质量。结果:建立了20批不同来源山楂叶的专属性指纹图谱,确定了15个共有峰;不同来源样品之间差异较大,通过OPLS-DA可将野生和市售山楂叶显著分成两类;经过对照品对照可知,酚酸类是其主要的差异标记物,可作为其质量控制指标。结论:该方法的验证结果表明所建立的方法具有较好的重复性,简便可靠,结合模式识别方法可用于山楂叶药材野生与市售的鉴别,发现绿原酸是引起野生与市售山楂叶差异的主要成分,该方法可为山楂叶的质量评价提供新的科学依据。Objective:To establish a fingerprint of wild grown and commercial leaves of Crataegus pinnatifida from different regions.Methods:The fingerprint of Crataegus pinnatifida leaves was carried out on a Waters ACQUITY UPLC BEH C18 column(100 mm × 2.1 mm,1.7 pm) at 30 ℃,with a gradient mobile phase composed of acetonitrile-0.1% aqueous formic acid,the flow rate was 0.3 mL/min,the detection wavelength was 355 nm.The quality assessment was achieved by using chemometrics methods including similarity analysis,principal component analysis (PCA)and orthogonal partial least squares discriminate analysis (OPLS-DA).Results:A specificity fingerprint chromatogram of 20 batches Crataegus pinnatifida leaves was established,and 15 common peaks were designated.The disparities between samples from different sources was large,and the wild grown and the commercial Crataegus pinnatifida leaves were divided into two categories by OPLS-DA method.Phenolic acids was the main difference between markers after standard comparison,which can be used as the quality control index.Conclusion:The method validation results demonstrated that the method is desirable reproducibility,simple and reliable,combining with chemical mode can be used for the identification and distinguishing of wild grown and commercial Crataegus pinnatifida leaves.Chlorogenic acid is the main component for distinguishing wild grown and commercial Crataegus pinnatifida leaves.This method can be used to provide a new scientific basis for the quality evaluation of Crataegus pinnatifida leaves.
