摘 要:目的:研究美洲大蠊提取物Ento-A对噁唑酮(OXZ)诱导大鼠溃疡性结肠炎(UC)的疗效,并初步探讨其作用机制。方法:将雄性SD大鼠分为正常对照组、模型组、美沙拉嗪组和美洲大蠊提取物Ento-A低、中、高剂量组,除正常对照组外,其余各组以噁唑酮溶液诱导大鼠溃疡性结肠炎模型。测定疾病活动指数(DAI)、脏器指数、结肠黏膜损伤指数(CMDI)和病理组织学评分(HS),检测血清IL-4、IL-10、iNOS及结肠黏膜中MPO、EGF水平,评价药物对UC大鼠的疗效。结果:与正常对照组比较,模型组血清IL-4、IL-10及结肠组织EGF水平显著降低,CMDI评分、HS评分、血清iNOS、结肠指数及结肠MPO活性显著升高。美沙拉嗪组及Ento-A各剂量组上述指标均得到不同程度改善。结论:美洲大蠊提取物Ento-A灌肠能有效缓解噁唑酮诱导的大鼠UC,其作用机制可能与下调iNOS、MPO水平,上调IL-4、IL-10及EGF水平有关。Objective:To study the therapeutic effect of Periplaneta americana extract Ento-A on oxazolone induced ulcerative colitis in rats,and to explore its mechanism.Methods:Male SD rats were divided into normal control group,model group,the mesalazine group and Ento-A low-,medium-and high-dose groups.In addition to the normal control group,other groups were induced into ulcerative colitis with oxazolone solution.The disease activity index(DAI),organ index,colon mucosa damage index (CMDI)and histopathological score(HS),the levels of IL-4,IL-10,iNOS in serum,and MPO and EGF in colonic mucosa were dected to evaluate the therapeutic effect of drug on ulcerative colitis in rats.Results:Compared with the normal group,the level of IL-4 and IL-10 in serum,EGF in colonic were reduced significantly,while the CMDI score,HS score,the content of serum iNOS,colon index and MPO in colonic were increased significantly in the model group.Abore indicators of mesalazine group and Ento-A low-,medium-and high-dose groups were with varying degrees of improvement.Conclusion:Periplaneta americana extract Ento-A can effectively alleviate oxazolone induced ulcerative colitis in rats,the mechanism may be related to the down regulation of iNOS,MPO levels and the up regulation of IL-4,IL-10,EGF levels.
