摘  要:目的:实验通过采集黄河三角洲地区耐盐植物盐芥的根际土壤,采用Martin法、SDS法、高盐改进法3种方法提取根际土壤的总DNA,测定提取的纯度和浓度。方法:对以上3种方法进行改进后,获得了较高的DNA质量和提取率。结果:采用SDS加蛋白酶K和溶菌酶的方法,提取总DNA的浓度为145.3μg/g,A260/A280为1.82;在高盐改进法中将样品反复冻融5次,提取总DNA的浓度为141.3μg/g,A260/A280为1.81。优化后的提取方法均得到了纯度较高的DNA样品,本实验为后续微生物多样性的研究提供了基础。Objective:Total DNA of the rhizosphere soil from the salt-tolerant plants Thellungiella salsugineain theYellow River Delta was extracted by Martin method,SDS method and improved high-salt method. Method:Two mod-ified methods were obtained. Conclusion:From the SDS method added with protease K and lysozyme,the concentra-tion of total DNA was 145.3μg/g,A260/A280 was 1.82. After 5 times of frozen-melt treatments,141.3μg/g totalDNA concentration was obtained using improved high-salt method,A260/A280 was 1.81. These two methods got pu-rified DNA samples,and provided the foundation for the further research of microbial diversity.
