摘  要:目的本研究对翅果油树(Elaeagnus mollis Diels)种子CO_2超临界萃取物(EMSC)进行成分分析,并初步筛选其抗炎及神经保护活性,旨在为其开发利用提供实验依据。方法采用CO_2超临界萃取技术对翅果油树种子进行萃取,并通过气相色谱-质谱联用仪(GC-MS)法检测主要化学成分,采用峰面积归一化法得出各组分的相对百分含量。以二甲苯致小鼠耳肿胀炎症模型和大鼠皮层神经元细胞氧糖剥夺损伤模型初步考察超临界萃取物(EMSC)抗炎及神经保护活性。结果翅果油树种子CO_2超临界萃取出油率为35.6%,通过GC-MS鉴定了4种脂肪酸成分,分别为棕榈酸、亚油酸、油酸和硬脂酸。EMSC低、中和高剂量组小鼠耳肿胀度与空白对照组小鼠耳肿胀度比较差异均有统计学意义(P<0.05);低、中和高剂量组小鼠耳肿胀的抑肿率分别为52.63%、65.27%和74.22%。EMSC在0.2~40μg/m L的剂量下均有显著的神经保护活性,且在0.5~10μg/m L剂量下细胞生存率均值可达80%以上。结论 EMSC在二甲苯致小鼠耳肿胀模型中表现出较好的抗炎活性,在大鼠皮层神经元细胞氧糖剥夺模型中具有显著的神经保护活性。Objective To analyzed the chemical constituents of SCF -CO2 extract from Elaeagnus mollis seeds (EMSC) by GC - MS. The anti - inflammatory and neuroprotective effect of EMSC were screened to provide experimental basis for its exploitation and utilization. Methods The seeds of Elaeagnus mollis were extracted by supercritical fluid CO2. The chemical profiles of extract were analyzed by gas chromatography and mass spectrometry (GC -MS), and the relative percentage content of each composition were calculated by peak area normalization method. The anti - inflammatory effect of EMSC were observed by the inflammation model on mouse ear swelling induced by xylene, and the neuroprotective effects of which were investigated by MTT assay in oxygen - glucose deprivation induced cortical neurons injury model. Results The yield of oil recipe of EMSC was 35.6%. Four fatty acid components were identified by GC -MS,which were palmitic acid,linoleic acid,oleic acid and stearic acid. The mouse ear edema in low,medium and high dose group of EMSC had significant difference compared with the blank control group (P 〈 0. 05 ). The swollen suppression rate for mouse? ear? edema in low,medium and high dose group were respectively 52.63% ,65.27% and 74.22%. The EMSC at dose of 5 -200 uM had significant protect effect in OGD - induced cortical neurons injury model compared with model group, and the cell viabilities were over 80% at dose of 10 - 80 μM. Conclusion The EMSC show better anti - inflammatory activity on the inflammation model in mouse ear swelling induced by xylene, and which can protect cortical neurons injured by oxygen -glucose deprivation model.
