健脾养血祛风方对慢性湿疹小鼠模型趋化因子CCL17和CCL18的影响

摘　　要：目的观察健脾养血祛风方对慢性湿疹小鼠模型血清及皮肤组织中趋化因子CCL17和CCL18的作用。方法取健康雄性小鼠50只,随机分为5组,即空白对照组、慢性湿疹模型组、慢性湿疹西药治疗组、慢性湿疹中药治疗高剂量组及低剂量组。中药组以健脾养血祛风方进行治疗,西药组以盐酸左西替利嗪治疗;采用流式细胞仪检测小鼠脾细胞CD4^+和CD8^+水平并计算其比值;采用ELISA法检测小鼠血清嗜酸性粒细胞活化趋化因子CCL17和CCL18水平;采用RT-PCR法检测小鼠背部皮肤组织中CCL17和CCL18的表达。结果治疗组干预后,小鼠脾细胞CD4^+水平、CD4^+/CD8^+显著升高,血清中CCL17和CCL18水平显著下降,小鼠皮损中CCL17和CCL18的表达明显减弱。以上与模型组相比差异均具有统计学意义(P<0.05)。中药高剂量组各指标与西药组比较差异均无统计学意义(P均>0.05)。结论健脾养血祛风方能够显著上调慢性湿疹小鼠模型CD4^+水平及CD4^+/CD8^+比值;下调慢性湿疹小鼠模型趋化因子CCL17及CCL18血清水平、抑制CCL17及CCL18在病变组织中的表达,对失衡状态下的细胞免疫具有较好的调节作用。Objective To determine the effects of Jianpi Yangxue Qufeng formula on CCL17 and CCL18 expression in serum and skin tissues of mice with chronic eczema. Methods Fifty healthy male mice were randomly divided into 5 groups,namely normal controls,chronic eczema controls,chronic eczema treated with western medicine（Levocetirizine Dihydrochloride）,and chronic eczema treated with high or low dose of Chinese herbs. Flow cytometry was used to measure expression levels of CD4＋ and CD8＋ in spleen cells. ELISA method was used to measure expression levels of CCL17 and CCL18 in serum,while expression levels of CCL17 and CCL18 mRNA in skin tissues were assessed with RT-PCR. Results Treatments with either western medicine or herbal medicines significantly increased the expression levels of CD4＋ and CD4＋/CD8＋ in spleen cells（P〈0.05 vs. untreated）,and decreased the expression levels of CCL17 and CCL18 in the serum（P〈0.01）. The expression levels of CCL17 and CCL18 in the skin were also significantly reduced after treatments with either western medicine or herbal medicines（P〈0.01）. The efficacy of Chinese herbs at high dose was comparable to that of Western medicine（P all〉0.05）. Conclusion Jianpi Yangxue Qufeng formula can increase the level of CD4＋ in spleen cells and the ratio of CD4＋/CD8＋,in addition to downregulation of CCL17 and CCL18 expression in the serum and the skin. Jianpi Yangxue Qufeng formula can regulate the imbalanced immune cells.
