摘  要:目的探讨天麻素对肌萎缩侧索硬化症h SOD1^(G93A) NSC34细胞模型的影响及机制。方法给予正常培养的NSC34细胞天麻素后,观察细胞形态活力变化。通过转染h SOD1^(WT)和h SOD1^(G93A)质粒,建立h SOD1^(WT)和h SOD1^(G93A)NSC34稳定细胞系后再给予天麻素干预以观察h SOD1^(WT)和h SOD1^(G93A) NSC34细胞形态和活力变化以及MDA水平。结果在正常培养的NSC34细胞内加入天麻素后,细胞的形态和活力均无明显影响(P>0.05);在成功转染h SOD1^(WT)和h SOD1^(G93A)质粒的NSC34细胞内加入天麻素后,细胞活力明显增加(P<0.05,P<0.01),MDA水平下降(P<0.01)。结论天麻素对正常培养的NSC34细胞无明显影响,但对转染h SOD1G93A质粒后的细胞,有明显促进细胞存活和降低MDA的作用。Objectives To investigate the mechanisms and effects of gastrodin on hSOD1^(G93A) NSC34 cell model of amyotrophic lateral sclerosis(ALS). Methods After treating with gastrodin, the change of cell morphology and viability of NSC34 cell that was normally cultured were accessed. The hSOD1^(WT) and hSOD1^(G93A) NSC34 cell models were esbalished via transfecting hSOD1^(WT) and hSOD1^(G93A) plasmids;and the change of cell morphology,viability and MDA level after treating with gastrodin were also observed. Results Treating with gastrodin did not significantly influence cell morphology and viability(P〈0.05). In cells transfected with hSOD1^(WT) and hSOD1^(G93A) plasmids,the cell viability was significantly elevated(P〈0.001), and MDA level was significantly increased(P〈0.01) after treating with gastrodin. Conclusion Treating with gastrodin does not have significant influence on normally cultured NSC34 cells,but could resist the cellular damage caused by hSOD1G93A plasmid transfection and promote cell survival via reducing MDA level.
