摘 要:目的观察木豆叶提取物对抗维甲酸导致大鼠骨质疏松的作用。方法取SD大鼠60只,雌雄各半,按体质量随机分为5组,即正常组,模型组,仙灵骨葆阳性药组(300 mg·kg^(-1)),木豆叶提取物高、低剂量组(8,0.8 g·kg^(-1))。除正常组外,每组大鼠均上午灌胃维甲酸14 d,造模同时各组分别给予相应剂量的药物进行干预,连续给药4周。观察木豆叶提取物对大鼠体质量、脏器指数以及骨钙、磷、骨骼灰化率和骨组织形态学的影响。结果与模型组比较,木豆叶提取物低剂量组能显著提高肾上腺和前列腺的指数(P<0.05),高剂量组能减少脾脏指数(P<0.05);木豆提取物高、低剂量组对股骨和椎骨骨钙、磷含量及骨骼灰化率都有显著提高作用(P<0.05,P<0.001);病理形态学指标结果显示,模型组动物皮质骨面积百分比、骨小梁平均厚度、骨小梁面积、骨小梁体积、成骨细胞数均较正常组明显减少(P<0.001),而破骨细胞数明显增多(P<0.001)。木豆叶提取物低剂量组能明显对抗上述指标中除成骨细胞数的变化(P<0.001),高剂量组对除骨小梁厚度和成骨细胞数之外的上述指标变化有逆转作用(P<0.05,P<0.01,P<0.001)。结论木豆叶提取物能较好地改善维甲酸造成的大鼠骨质疏松。Objective To study the impact of the extracts of Cajanus cajan L.(ECC) on retinoic acid induced osteoporosis in rats. Methods A total of 60 Sprague-Dawley rats with half males and half females were randomly devided into five groups according to their weight:normal control group,osteoporosis model group,ECC(8 g·kg^(-1))treatment group,ECC(0.8 g·kg^(-1)) treatment group and Xianling Gubao postive control group. Except the normal control group, all groups were orally administrated with retinoic acid daily, continuously for 14 days. At the same time, each group was given the corresponding drug to intervene for 28 days. The effect of ECC on weight, viscera index, bone calcium, bone phosphorus contents, bone ashing rate and bone tissue morphology in rats with osteoporosis induced by retinoic acid were observed. Results Compared with the model group, the low dose of ECC significantly increased adrenal and and prostate index(P〈0.05); and the spleen index was significantly decreased after the rats treating with high dose of ECC(P〈0.05). ECC could significantly increased calcium,phosphorus and ashing rate of femur and vertebrae of bone(P〈0.05,P 0.001). The results of morphological examination showed that the cortical bone area percentage, thickness,area percentage and volume of trabecular bone,and the number of osteoblast of the osteoporosis model group were significantly decreased compared with the normal control group(P 〈0.001), while the number of osteoclast markedly increased(P〈 0.001). Changes of the above indicators except number of osteoblast were reduced after the rats treated with low dose of ECC(P 〈0.001). In addition,changes of the above indicators except number of osteoblast and thickness of trabecular bone were reversed after the rats treated with high dose of ECC(P〈0.05,P〈0.01,P〈 0.001). Conclusion ECC had preventive effect on retinoic acid-induced osteoporosis in rats.
