摘　　要：目的 :为临床使用naldemedine提供用药参考。方法:从naldemedine的作用机制、药代动力学、药效学、临床研究及药物安全性等方面进行阐述。结果:Naldemedine可显著增加便秘患者的排便次数,对疼痛无影响且不会引起阿片戒断症状。结论:Naldemedine是一种口服的阿片受体拮抗药,主要用于治疗成人慢性非癌性疼痛患者中阿片类药物引起的便秘,为便秘患者提供了新的治疗手段。但是尽管该药已成功在美国获批上市,但其在不同地域及不同种族患者中的疗效仍需进一步的研究。Objective：To provide reference for rational use of naldemedine.Methods：The mechanism of action,pharmacokinetics,pharmacodynamics,clinical studies and safety of naldemedine were described.Results：Naldemedine can significantly increase the defecation frequency of patients with opioid-induced constipation.It has no effect on pain and does not cause opioid withdrawal symptoms.Conclusion：Naldemedine is an oral opioid antagonist,which is mainly used for the treatment of opioid-induced constipation in adult patients with chronic non-cancer pain.Naldemedine provides a new means of treatment for opioid-induced constipation.Although the drug has been successfully approved in the United States,but further research is still necessary to verify its efficacy for patients of different races in different regions.
