尿路上皮癌抗原1在子宫内膜异位症患者在位内膜中的表达及意义

摘　　要：目的检测子宫内膜异位症患者在位内膜中lncRNA尿路上皮癌抗原1(UCA1)的表达情况,探讨其在子宫内膜异位症病因学中的意义。方法选取2015年3-11月在中国医科大学附属盛京医院经病理证实为子宫内膜异位症患者的在位内膜标本30例(其中分泌期13例,增生期17例)为实验组,正常子宫内膜标本20例(分泌期7例,增生期13例)为对照组。采用实时荧光定量聚合酶链式反应(qRT-PCR)技术检测两组子宫内膜组织中UCA1的表达。结果实验组在位内膜、增生期内膜和分泌期内膜中UCA1相对表达量明显高于相同时期对照组,差异均有统计学意义(均P<0.01)。而两组组内增生期与分泌期的子宫内膜中UCA1的相对表达量比较,差异均无统计学意义(均P>0.05)。结论在位内膜中UCA1基因的高表达与子宫内膜异位症密切相关,可能参与子宫内膜异位症的发生发展。UCA1的表达水平可能不受内膜月经周期变化的影响。Objective To detect the expression of lncRNA urothelial carcinoma associated antigen 1（UCA1） in eutopic endometrium of patients with endometrosis,explore the significance in etiology of endometriosis.Methods Thirty samples of eutopic endometrium from endometriosis patients confirmed by pathological examination in the hospital from March to November in 2015 were selected as experimental group,including 30 samples during secretory phase and 17 samples during proliferative stage;twenty samples of normal endometrium were selected as control group,including 7 samples during secretory phase and 13 samples during proliferative stage.Real-time fluorescent quantitative PCR（qRT-PCR） was used to detect the expressions of UCA1 in the two groups.Results The relative expression levels of UCA1 in eutopic endometrium,endometrium during proliferative phase,endometrium during secretory phase in experimental group were statistically significantly higher than those in control group（all P〈0.01）.In the two groups,there was no statistically significant difference in the relative expression levels of UCA1 in endometrium between proliferative phase and secretory phase（all P〈0.05）.Conclusion High expression of UCA1 in eutopic endometrium may be closely correlated with endometriosis,UCA1 may be involved in occurrence and development of endometriosis.The expression level of UCA1 may not be affected by change of menstrual cycle of endometrium.
