和胃降逆方对非糜烂性胃食管反流病大鼠5-羟色胺及其受体表达的影响(一)

摘　　要：目的:观察和胃降逆方对非糜烂性胃食管反流病(NERD)大鼠5-羟色胺(5-hydroxy tryptamine,5-HT)及其受体表达的影响,探讨和胃降逆方对非糜烂性胃食管反流病大鼠的作用机制。方法:将SPF级SD大鼠随机分为空白组、模型组、奥美拉唑组、和胃降逆方高剂量组、和胃降逆方中剂量组、和胃降逆方低剂量组,采用基础致敏联合酸灌注的方法建立内脏高敏感NERD模型;干预2周后取血清与食管组织;ELISA法测定大鼠血清及食管中5-HT的含量,HE染色评价大鼠食管病理改变情况,免疫组织化学法测定5-HT3R与5-HT4R的表达水平。结果:和胃降逆方中、低剂量组、奥美拉唑组大鼠血清5-HT含量均低于模型组(P<0.05);和胃降逆方中剂量组、奥美拉唑组大鼠食管5-HT含量低于模型组(P<0.05);和胃降逆方高、中剂量组大鼠食管5-HT3R表达量明显低于模型组(P<0.05)。结论:和胃降逆方可降低NERD大鼠血清及食管5-HT水平,其作用机制可能与降低食管5-HT3R的表达有关。Objective： To observe the effects of HWJNF on 5-hydroxytryptamine and receptors of non-ero- sive gastroesophageal reflux disease rats and to investigate mechanism of HWJNF. Methods： The SD rats were randomly divided into 6 groups, control group, model group, omeprazole group and the high, medium and low groups of HWJNF; Except the control group, the rats of all the other groups received basal ovalbumin-sensiti- zation combined with intra-esophageal mucosal acid exposure for making NERD models. The treatment was giv- en for 2 weeks. The comparative observation was carried out on the changes of pathologic structure and the expression of 5-HT, 5-HT3R, 5-HT4R via the methods of Elisa, HE and immunohistochemical staining. Re- suits： The middle and low groups of HWJNF and omeprazole group showed lower expressive levels of 5-HT in the serum than the model group （P〈0.05）, besides, the middle group of HWJNF and omeprazole group showed lower expressive levels of 5-HT in the esophageal than the model group （P〈0.05）. The high and medium groups of HWJNF showed lower levels of 5-HT3R than the model group （P〈0.05）. Conclusion： HWJNF can reduce the levels of 5-HT in serum and esophageal, the mechanism of which may be in relation with decreasing the level of 5-HT3R in the esophageal.
