摘 要:目的:探讨健脾益肠散对实验性溃疡性结肠炎(UC)大鼠细胞间黏附分子1(ICAM-1)的影响。方法:将健康SPF级雄性SD大鼠60只,随机分为正常组和造模组;造模组采用TNBS/乙醇法复制UC大鼠模型;待复制模型成功后将造模组随机分为5组,分别为模型组、西药组及中药高、中、低剂量组,每组10只;灌胃相应药物3周后,HE染色检测各组大鼠结肠黏膜组织损伤情况,同时采用ELISA法检测血清中ICAM-1的水平,免疫组化法检测结肠组织中ICAM-1的表达。结果:模型组与正常组相比,大鼠结肠黏膜损伤评分、血清ICAM-1水平及结肠组织中ICAM-1表达均明显升高(P<0.01)。中药高、中剂量组血清ICAM-1水平和高、中、低剂量组结肠黏膜损伤评分、ICAM-1表达均较模型组下降(P<0.05或P<0.01)。中药中剂量组对ICAM-1的影响与西药组比较,差异无统计学意义(P>0.05)。结论:健脾益肠散可能通过调节ICAM-1而达到对UC大鼠肠黏膜的免疫保护,从而发挥治疗作用。Objective: To explore the changes of Intercellular adhesion molecule-1 (ICAM-I) in ulcerative enlitis (UC) rats and the effect of Jianpi Yichang Powder on it. Methods: A total of 60 healthy SPF male SI) rats were randomly divided into normal group and model group. The UC models were established by TNBS/ ethanol method. Then the UC model rats were randomly divided into the model group, western medicine group, and TCM high, middle and low dose group with 10 rats in each. After gavage for 3 weeks, HE staining was used to observe the injury of colonic mueosa; ELISA method was used to detecl the content of ICAM-I in serum and the protein expression of 1CAM-I in colon was detected by immunohistoehemistU. Results: Compared with the normal group, the colon mueosal injuU score, serum ICAM-1 content and ICAM-I protein expression were increased significantly in model group (P〈0.01). The TCM high, middle-dose group showed lower serum levels of ICAM-1 than the model group and the TCM high, middle, lnw-dose group showed lower cohm mucosal injury score and ICAM-I protein expression than the model group (P〈0.05 or P〈0.01). Effect of TCM middle- dose group on ICAM-I with western medicine group was not significant (P〉0.05). Conclusion: Jianpi Yiehang Powder can regulate the ICAM-I to protect intestinal mucosal immunity of UC rats as to play a therapeutic role.
