摘  要:目的:探讨桑根酮C对小鼠急性炎症反应的作用及其机制。方法:昆明小鼠随机分成6组,每组10只,分别腹腔注射桑根酮C 5、15、50、100 mg/kg、地塞米松5 mk/kg(阳性对照组)、相同体积的二甲基亚砜(DMSO,溶剂组),给药30 min后,各鼠腹腔注射0.6%醋酸0.2 m L/只,30 min后处死小鼠,剖腹收集腹腔洗液,应用紫外分光光度计测上清液吸光度A值分析桑根酮C对醋酸所致小鼠腹腔毛细血管通透性增高的影响。6组给药30 min后观察桑根酮C对角叉菜胶所致小鼠足肿胀的影响,在各鼠右后足跖皮下注射1%角叉菜胶30μL致炎,5 h后处死小鼠,对称剪下鼠足计算肿胀度。然后去皮剪碎后离心取上清液,用考马斯亮蓝法测蛋白的含量、ELISA法测定足跖组织中各种炎症反应因子髓过氧化物酶(MPO)、NO、前列腺素2(PGE2)、白细胞介素1β(IL-1β)及肿瘤坏死因子(TNFα)的水平。结果:桑根酮C可抑制醋酸所致小鼠腹腔毛细血管通透性增加和抑制角叉菜胶引起的小鼠足肿胀,溶剂组A值和肿胀度均高于阳性对照组[0.693±0.101比0.491±0.051,(78.37±6.55)mg比(39.52±5.11)mg,均P<0.05],桑根酮C各剂量组A值和小鼠足肿胀度均低于溶剂组(均P<0.05),且随剂量的增加A值和足肿胀度降低明显,相关分析显示桑根酮C对醋酸致小鼠腹腔毛细血管通透性增加和角叉菜胶所致小鼠足肿胀的抑制作用呈剂量效应关系(r=0.761,P=0.013;r=0.794,P=0.008)。桑根酮C各剂量组和阳性对照组能明显降低炎症反应渗出液中蛋白及MPO、NO、PGE2、TNFα及IL-1β的含量,各组各指标含量均低于溶剂组(均P<0.05),且随桑根酮C抑制各炎症反应因子的释放呈剂量效应关系。结论:桑根酮C通过减少中性粒细胞浸润及抑制各种炎症反应介质的生成和释放来发挥抗炎作用。Objective:To investigate the effect of Sanguonone C on acute inflammatory response in mice and its mechanism. Methods:The Kunming mice were randomly divided into 6 groups ( n=10 each ) , and were intraperitoneally injected with 5,15,50 and 100 mg/kg Sanguonone C,5 mk/kg Dexamethasone ( positive control group),and the same volume of dimethylsulfoxide (DMSO,solvent group),respectively. At 30 min after the administration,the mice were injected with 0. 6% acetic acid (0.2 mL each). At 30 min,the mice were sacrificed and the peritoneal lavage fluid was collected in the laparotomy. The UV spectrophotometer was used to measure the absorbance A value of the supernatant. The effect of Sanguonone C on the acetic acid-induced increase of capillary permeability in mice was analyzed. At 30 min after the administration,the effect of Sanguonone C on feet swelling induced by Carrageenan was determined. The mice received subcutaneous injection of 30μL 1%Carrageenan at the right hind paw. At 5 h after the mice were sacrificed,the mice feet were cut symmetrically to calculate the degree of swelling. After the skin was removed,the tissues were cut into pieces and centrifuged to obtain the supernatant. The protein content was measured by Coomassie brilliant blue method. The levels of inflammatory response factors,myeloperoxidase (MPO),NO,prostaglandin 2 (PGE2),interleukin-1β (IL-1β) and tumor necrosis factor ( TNFα) in the paw tissues were measured by ELISA. Results:Sanggenon C could inhibit the increase of capillary permeability induced by acetic acid, and inhibit the feet swelling caused by Carrageenan. The A value and swelling degree in the solvent group were higher than those in the positive control group [0.693&#177;0.101 vs 0.491&#177;0.051,(78.37&#177;6.55) mg vs (39.52&#177;5.11) mg,all P〈0.05].
