摘 要:目的探讨促红细胞生成素对急性心肌梗死(AMI)大鼠心肌核转录因子-κB(NF-κB)与转录因子活化蛋白-1(AP-1)蛋白表达的影响。方法将36只健康雄性SD大鼠随机分为假手术组、急性心肌梗死组和促红细胞生成素(EPO)治疗组。急性心肌梗死组及EPO治疗组通过冠状动脉左前降支结扎建立急性心梗模型。模型建立4周后,利用心脏超声评价心功能,采用免疫组织化学方法与Western blot方法检测各组大鼠心梗区域心肌NF-κB与AP-1蛋白的表达。结果与急性心肌梗死组大鼠相比,EPO治疗组大鼠心功能明显改善,左室射血分数明显增加,左室舒张末期内径(LVEDD)明显缩小(P<0.01)。急性心肌梗死组大鼠心肌NF-κB与AP-1蛋白表达量比假手术组显著升高,而EPO治疗组显著低于急性心肌梗死组(P<0.01)。结论降低NF-κB与AP-1蛋白表达可能是促红细胞生成素抑制急性心肌梗死大鼠炎性反应的机制之一。Objective To investigate the effects of erythropoietin (EPO) on the expression of nuclear factor kappa B(NF—κB) and transcription factor activator protein -I(AP-1) in myocardium of rats with acute myocardial infarction(AMI). Methods 36 healthy male SD rats were randomly divided into sham group, acute myocardial infarction group and erythropoietin (EPO) treatment group. An acute myocardial infarction model was established by ligation of the left anterior descending coronary artery. 4 weeks after the establishment of the model, cardiac function was evaluated by echocardiography, immunohistochemistry method and Western blot method were used to detect the expression of HIF- α and Survivin proteins in myocardium. Results Compared with the acute myocardial infarction group, the heart function was improved, the left ventricular ejection fraction was increased significantly, and the left ventricular end diastolic diameter(LVEDD) was reduced significantly in EPO treatment group (P〈0.01). The expression levels of NF-κ B and AP-1 proteins in myocardium were increased significantly in acute myocardial infarction rats than in sham group, but decreased significantly in EPO treatment group than in acute myocardial infarction gronp(P〈0.01). Conclusion The inhibition of erythropoietin on the inflammatory response in rats with acute myocardial infarction might be related to downregulation of NF- κB and AP-1 expressions.
