摘 要:目的:观察左归丸对雷公藤多苷(GTW)诱导的少弱精子症大鼠模型的精子参数、睾丸/附睾系数、生殖激素水平、附睾肉毒碱含量的预防作用。方法:将36只雄性SD大鼠随机平均分为正常组、模型组及左归丸组,每组12只,共计8周。模型组前4周用去离子水进行干预,5~8周用40mg/(kg·d)的雷公藤多苷进行造模;左归丸治疗组前4周用6g/(kg·d)的左归丸混悬液进行预防性干预,5~8周用40mg/(kg·d)的雷公藤多苷进行干预。检测大鼠精子参数;测量睾丸的重量,计算睾丸脏器系数;测定血清黄体生成素(LH)、卵泡刺激素(FSH)、雌激素(E_2)、泌乳素(PRL)、睾酮(T)、抑制素B(INHB)及附睾肉毒碱(EC)含量。结果:左归丸组与模型组相比,大鼠睾丸脏器系数、精子质量(活力、数量)、血清FSH、LH、T、INHB、EC含量方面的差异具有统计学意义(P<0.05或P<0.01);但INHB、EC水平仍与正常组有一定差异(P<0.05)。结论:前期应用左归丸能够有效减少GTW对大鼠生殖器官的损伤程度,包括对一般状态、睾丸/附睾脏器指数、精子质量及生殖激素水平、附睾肉毒碱含量的影响,具有一定的预防作用。Objectives: To observe the preventive effect of Zuogui Pill on sperm parameters,testis/epididymal coefficient,reproductive hormone level and carnitine content in epididymis of rats with oligoasthenospermia induced by tripterygium wilfordii( GTW). Methods: 36 male Sprague-Dawley rats were randomly divided into normal group,model group and Zuogui pill group,with 12 rats in each group,for a total of 8 weeks. The rats in model group were treated with deionized water for the first 4 weeks,and then the rats were treated with 40 mg/( kg·d)Tripterygium wilfordii for 5-8 weeks. The rats in Zuogui pill group were treated with 6 g/( kg·d) Zuogui pill for preventive intervention for the first 4 weeks,and 40 mg/( kg·d) of the tripterygium glycosides to intervene for 5-8 weeks.( LH),follicle-stimulating hormone( FSH),estrogen( E2),prolactin( PRL) and testosterone( T)were measured,and the testis weight was measured. Inhibin B( INHB) and epididymal carnitine( EC) were measured. Results: The differences in contents of FSH,LH,T,INHB and EC in testis between Zuogui pill group and model group were significantly different( P〈0. 05 or P〈0. 01),and the levels of INHB and EC in Zuogui pill group and model group were still different from these of normal group( P〈0. 05). Conclusion: Zuogui Pill can effectively reduce the damage degree of GTW on the reproductive organs of rats,including the effects of general state,testis/epididymis organ index,sperm quality and reproductive hormone levels,and the content of carnitine in epididymis. It is proved that Zuogui Pill has certain preventive effect on rats with oligoasthenospermia.
